摘 要:目的:验证核因子κB(nuclearfactor-kappaB,NF-κB)信号转导途径在骨性关节炎软骨细胞中被激活的假说。方法:实验于2005-07/09在南京大学生命科学院动物实验室完成。选取SPF级SD大鼠20只,均取左膝为骨性关节炎模型组,右膝为空白对照组。①骨性关节炎模型组行前交叉韧带切断术建立模型,术毕腹腔注射丁胺卡那(20mg/只)预防感染,连用3d。空白对照组不做处理。②术后6周取材。取膝前正中切口,离断关节周围肌肉,在股骨髁上剪断股骨,平台下方1cm剪断胫骨,取下游离的膝关节,剪断关节囊和内外侧韧带分开股骨髁和胫骨端,胫骨进行病理学实验,股骨进行分子生物学实验。将胫骨标本投入0.5mol/L的乙二胺四乙酸中脱钙8周,制作5μm切片,利用苏木精-伊红及甲苯胺蓝染色观察胫骨关节面的破坏程度,以Western-blot免疫印迹分析定量检测股骨髁软骨细胞NF-κB信号转导途径的激活程度(p65灰度积分)。结果:实验选取SD大鼠20只,全部进入结果分析。①术后6周两组关节软骨组织病理学检查结果:苏木精-伊红染色及甲苯胺蓝染色显示,空白对照组关节软骨表面平整,骨性关节炎模型组关节软骨表面破坏,粗糙不平。②两组p65条带灰度积分计算结果的比较:骨性关节炎模型组的灰度积分明显高于空白对照组(0.832±0.112,0.372±0.081,P<0.05)。结论:NF-κB信号转导途径在大鼠实验性骨性关节炎软骨细胞中被激活。ATM: To verify the hypothesis that nuclear factor-kappa B (NF-κB) signal transduction pathway is activated in osteoarthritis chondrocytes. METHODS: The experiment was carried out in the Animal Experiment Center of Life Science School, Nanjing University from July to September 2005. Twenty SPF SD rats were selected with the left knee as experiment group and the right knee as control group. ①The osteoarthritis models were established by anterior cruciate ligament transection, and then Amikacin intraperitoneal injection (20 mg per rat) was given for 3 days to prevent inflammation. The control group was not given any treatment. ②After 6 weeks of operation, all muscles around knee was cut off following anterior middle incision to cut down the femur above condyles, then the tibia was cut down 1 cm below tibial plateau. The articular capsule, lateral and medial collateral ligament were cut off to separate the femur and tibia. The proximal tibia was sent for pathological experiments and the distal femur was sent for molecular biological detections. The proximal tibia was put into 0.5 mol/L edathamil for decalcification for 8 weeks, and 5 μm pathological section was prepared. HE stain and toluidine blue stain were used to observe the damage degree of tibial cartilage. The activating level of femoral condyles chondrocytes NF-κB signal transduction pathway (p65 gray integral) was detected by Western-blot. RESULTS: All 20 rats entered the result analysis. ①Pathological results after 6 weeks of operation: The HE stain and toluidine blue stain showed that the joint surface of the control group was even, while it was seriously destroyed in the experiment group. ②Comparison of results of the two p65 gray integrals: NF-κB p65 in the experiment group was higher expressed than in the control group (0.832±0.112, 0.372±0.081, P 〈 0.05). CONCLUSION: NF-κB signal transduction pathway is activated in experimental osteoarthritis chondrocytes of rats.
