摘 要:应用RAPD标记方法对30个水稻栽培品种进行了籼粳分类研究。从80个随机引物中筛选出了扩增出具有多态性的谱带的18个引物,并从中选出重复性高、稳定性好的S1004,S1012,S1019,S1048,S1049,S1053,S1063,S1070,S1075共9个引物进行反应。依据随机引物对不同品种水稻总DNA扩增出的特异性扩增条带,用Ne i’s公式计算30个水稻品种间的相似系数,并以UPMGA法进行聚类分析,其结果与品种系谱基本吻合,证明RAPD标记可应用于水稻的分类。The present study used RAPD markers to study the relation of 30 rice cultivars. In the course of PCR, 9 primers were selected from 80 primers to screen the samples. These 9 primers have polymorphism, they are S1004 , S1012, S1019 , S1048 , S1049 , S1053 , S1063 , S1070 , S1071. Based on the pair wise comparisons of amplification products the genetic distance was calculated using Nei genetic distance and a dendrogram was constructed using an unweighted pair group method with arithmetical averages( UPGMA). These 30 rice cultivars were divided into 2 groups. This result was identical with pedigree analysis and proved that RAPD markers could be used in classification of rice cultivars.
