摘 要:目的研究厚朴酚(Magnolol,Mag)对脑缺血再灌注损伤的保护作用及其可能机制。方法采用腹腔注射厚朴酚(25、50、100mg/kg,共3 d)预处理后,通过线栓法制备大鼠右侧大脑中动脉栓塞缺血再灌注模型,于脑缺血2h再灌注24 h时,观察不同剂量厚朴酚对脑缺血再灌注大鼠的神经功能状态、脑梗死体积、脑含水量、脑组织髓过氧化物酶(myeloperoxidase,MPO)活性及脑组织肿瘤坏死因子-α(tumor necrosis factor-,αTNF-α)、白介素-1β(in-terleuk in-1,βIL-1β)含量的影响。结果厚朴酚预处理后,可明显改善大鼠神经病学症状,降低脑水肿程度,降低脑梗死体积;可降低缺血再灌注后脑组织MPO活性,降低脑组织TNF-α、IL-1β的含量。结论厚朴酚对脑缺血再灌注神经元损伤具有良好保护作用,其机制可能与其抗炎作用有关。Objective To study the protective effects of magnolol on cerebral ischemia - reperfusion injury in rats and explore the mechanisms. Methods Magnolol (25,50 or 100 mg/kg/d,3 days) was given through intraperitoneal injection. Focal cerebral ischemia - reperfusion model was established in rats through occluding the middle cerebral artery(MCAO) on right side. The protective effects of magnolol at different doses was evaluated by investigating neurological score, infract size of brain, cerebral water contents, myeloperoxidase(MPO) activity ,the contents of tumor necrosis factor -α( TN F -α) and interleukin -1β ( I L -1β). Results ①Magnolol not only obviously alleviated the neurological symptoms, but also reduced the infract size and water contents. ②Magnolol decreased the MPO activity in brain tissue.③Magnolol decreased the contents of TNF-α and IL-1β in brain tissue. Conclusion Magnolol showed obvious protective effects on focal cerebral ischemia - reperfusion injury in rat and the protective effects might work through the anti -inflammation activity of Magnolol.
