摘  要:目的研究细胞间粘附分子(ICAM1)在鼠脊髓再灌注损伤过程中表达变化规律。方法将Wister大鼠随机分为实验组和对照组,建立脊髓再灌注损伤动物模型。采用逆转录-聚合酶链反应、免疫组化及免疫荧光激光共聚焦扫描显微镜技术检测脊髓再灌注损伤血管内皮ICAM1mRNA表达量。结果正常组和单纯缺血组不引起黏附分子表达量的增加(P>0.05)。而再灌注后缺血区黏附分子的表达及PMN的浸润先后发生了改变;再灌注后ICAM1呈递增性表达。再灌注2h组,ICAM1mRNA的表达量为(0.94±0.12)V,约为对照组2倍(P<0.05)。再灌注6h达到高峰,并持续到12h达高峰。再灌注12h,其在单位微血管面积上的荧光强度约比单纯缺血组增加了1/2(P<0.01)。结论细胞间黏附分子1在脊髓再灌注损伤过程中递增规律性表达,可能是促进脊髓再灌注损伤炎症进展重要病理因素。Objective To investigate expression and its Change Regularity of intercellular adhesion molecule-1 (ICAM-1)on the spinal cord repeffusion injury. Methods To set up the model of spinal cord reperfusion. To check the amount of the expression of ICAM-1mRNA of vascular endothelium of spinal cord repeffusion injury through the RT-PCR. immunohistochemistry, immunfluorescence and the technique of the Laser confocal Scanning Microscope. Results The amount of the expreesion of adhesion molecules does not increase in the normal group and the isehemia group. ( P 〉 0.05), while the amount of the expression of adhesion molecules in the ischemia area after reperfusion were appear changes of increase by degrees.The amount of the expression of ICAM-lmRNA increases to(0.94 ± 0.12)V after two-hour reperfusion, which is almost twice as much as that of the comparative groups( P 〈 0.05). It reaches the peak after six-hour reperfusion, and lasts twelve hours. After twelve-hour reperfusion its fluorescent intensity on per microvaseular area increases by one half more than that of the isehemia group. Condusion After the reperfusion injury, expression regularity of increase by degress of ICAM-1 were might an important pathology factor that the it would be promote inflammation progress of thc spinal repeffusion injury.
