摘  要:目的探讨粒细胞集落刺激因子(G-CSF)对高原急性多脏器功能不全综合征(M ODS)大鼠心肌的保护作用。方法在马汉山梯度(海拔3 900 m)建立20只大鼠M ODS模型,观察组(10只)造模前皮下注射G-CSF 10μg/(kg.d),连用5 d;对照组不用药。造模后24 h处死两组大鼠,取心肌标本,测定心肌梗死面积、左心室收缩及舒张期功能、心肌线粒体三磷酸腺苷(ATP)含量及F as mRNA、caspase-3的表达。结果观察组较对照组心肌收缩及舒张期功能增高明显,心肌ATP含量增加,心肌细胞凋亡数量减少,心肌中F as mRNA、caspase-3表达下降,心肌梗死面积缩小。结论G-CSF可提高高海拔地区M ODS大鼠心功能,其机制可能为动员骨髓基质干细胞修复坏死的心肌组织。[Objective]To study treatment on myocardium in multiple organ dysfunction syndrome rat at high altitude through Granulocyte Colony Stimulating Factor (G-CSF) . [Methods]The two-hit model of MODS rat was used at high altitude (3900m). Before set up the model, G-CSF was administrated by Subcutaneous injection. Hemorrhage was induced in Wistar rat by catheterizing the femoral artery and bleeding until a mean arterial pressure = 35mmHg was reached and maintained for 1 hour. Rususcitation was performed was with lactated Ringer? s. Cecal ligation and puncture(CLP) was performed 24 hours after hemorrhage. Rat was killed. Isolated hearts from male Wista rat weremounted on a Longendorff apparatus to estimate left ventricular systolic and diastolic function at different time points. The expression of Fas mRNA and the activity of Caspase-3 in myocardium was detected. Frozen myocardium was analyzed concentration of adenosine triphosphate. Myocardial infarct size was measured from mid-transverse sections stained with Masson's trichrome. [Results]Compared without G-CSF, the G-CSF can improved the left ventrlcular function and the concentration of myocardium adenosine triphosphate at high altitude more significant (P'~0. 05), the expression of Fas and Caspase-3 in myocardium at high altitude was lower (P〈0.05) and the myocardial infarct size was reduced. [Conclusion] Through applying the G-CSF, the marrow stromal cells moved and repaired the myocardium in MODS rat at high altitude. Our finding indicate that infusion G-CSF may represent a promising therapeutic strategy on myocardium of MODS rat at high altitude.
