摘  要:目的 制定柴胡药材中柴胡皂苷a、d的含量测定方法。方法 用高效液相色谱-蒸发光散射检测法(HPLC-ELSD),Alltima C18色谱柱,流动相甲醇-水(80:20),速度0.8ml·min^-1;蒸发光散射检测器参数:漂移管温度40℃,气压3.5bar。结果柴胡皂苷a和柴胡皂苷d分别在1.908-19.08μg(r=0.9996)和1.804-18.04μg(r=0.9992)范围内线性关系良好,平均回收率分别为98.35%(RSD=1.26%)和97.64%(RSD=1.58%)。结论 该方法简便、灵敏、可靠、准确、重现性好,可作为柴胡药材的质量控制方法。Objective To determine saikosaponin a, d in Radix Bupleuri by HPLC-ELSD. Methods HPLC-ELSD was used in the quantitative analysis by using Alhima CI8 chromatography column and methanol-water( 80: 20) as a mobile phase. The flow rate of mobile phase was 0. 8 ml · min^-1. The tube temperature of the detector was 40℃. The press of pure air was 3.5bar. Results The linear ranges for saikosaponin a and d were 1. 908 - 19.08 μg ( r = 0. 999 6 ) and 1. 804 - 18.04μg ( r = 0. 999 2 ) , the average recoveries were 98.35% ( RSD = 1.26% ) and 97.64% ( RSD = 1.58% ) , respectively. Conclusion This method is easy, sensitive , reliable, accurate , reproducible and can be used as the quality control method of Radix Bupleuri.
