摘 要:研究了从桑叶中提取分离1-脱氧野尻霉素(DNJ),探讨了提取剂、提取方法、料液比、提取次数、时间等因素对提取率的影响。实验表明:提取的最佳条件为以65%的乙醇为提取剂,按料液比1:8和1:4超声强化细胞破碎20min(功率1800W),732H阳离子树脂分离,0.25mol/L的氨水洗脱(洗脱速度1~2mL/min),提取率为95.8%,得率为124.54mg/100g,纯度为92.3%以上。本方法具有工艺流程简单、快速、提取率高的优点,适合于工业生产。The extraction and separation of 1-deoxynojirimycin (DNJ) from mulberry leaves was studied in this paper. The factors affecting the extracting yields such as solvent, method, the ratio of solid to liquid ,the time of extraction and the extracting times have been studied experimentally. The experiment shows that the optimal result of extraction is obtained when 65% alcohol and solution( 1:4 and 1:8) are used each once in ultrasonic extraction, and each extraction last 20 min power at 1800 W ,filtrate is separated by 732 H type resin cation,cleansed by ammonia water( 1~2 mL/min) ,in this way,the extraction ratio was 95.8% ,the yield could attain 124.54 mg/100 g, the purity exceeded 92.3%. This method is advantageous for its simple technical process, high speed and extraction rate. It is suitable for the industrialization.
