摘 要:目的跟踪观察机采浓缩血小板(platelet apheresis concentrates,PCs)在5%DMSO条件下于-80℃冻存1~5年的体外功能变化。方法玻璃珠柱检测血小板黏附功能,花生四烯酸诱导后检测血小板聚集功能,蝰蛇毒检测血小板Ⅲ因子活性。结果对深低温保存的冻存血小板的动态观察发现:PCs冻存期为1~5年融化复苏后的黏附和聚集功能未发现有统计学意义(P>0.05);冻存至第4年的血小板复苏时4/10样品发生凝集,第5年的有3/11发生凝集;冻存至第5年时,多数样品血小板Ⅲ因子失活。对22℃常温保存的液体血小板的观察发现:血小板的黏附功能和最大聚集能力随保存时间的延长逐渐下降,72h有差异(P<0.05),96h有显著性差异(P<0.01);5d之内血小板Ⅲ因子活性无明显改变(P>0.05)。结论22℃保存的液体血小板发生渐进的代谢损伤,而-80℃保存期在3年内的冻存血小板未见明显异常。Obieotive To follow up in vitro functional changes of apheresis platelet concentrates (APCs) cryo-preserved at -80 ℃ with 5% dimethylsulphoxide during a period from one to five years. Methods The platelet adhesion, aggregation and activity of factor m were tested by means of glass-ball column, abduction of arachidonic acid as well as vipe benom respectively. Results When the APCs were resuscitated from deep low temperature, the samples showed no significant changes in functions of adhesion and aggregation as the cryopreservation was prolonged from one to five years ( P 〉 0. 05 ). However, during the thawing process of the APCs, four out of ten and three out of eleven APCs samples appeared aggregative reaction after cryo-preservance for 4 and 5 years, respectively. The majority of the samples extensively lost their factor In activities in the fifth year storage. When fresh, liquid platelets were stored at room temperature (22 ℃ ), the in vitro adhesion and agglutination gradually decreased and statistic differences were observed in 72 h ( P 〈 0. 05 ) and in 96 h ( P 〈 0.01 ). However, the activity of factor Ⅲ was retained in the room temperature up to five days. Conclusion The liquid platelets stored at 22℃ were increasingly brought on metabolic injury, while the cry-oplatelets frozen at -80 ℃ in three years showed no evident changes.
