摘 要:以芝麻菜(Eruca sativaM ill)无菌苗的下胚轴、子叶和子叶柄作为外植体,研究了不同的激素浓度和组合对其愈伤组织诱导和植株再生的影响。结果表明,分别以MS+0.8 mg/L 2,4-D,MS+0.8 mg/L 2,4-D+0.1 mg/L6-BA,MS+0.8 mg/L 2,4-D+0.2 mg/L 6-BA为下胚轴、子叶柄和子叶的愈伤组织诱导培养基时,效果较好,出愈率依次为86.3%,78.2%,97.6%;将下胚轴、子叶柄和子叶的愈伤组织分别转移到分化培养基MS+3.0 mg/L6-BA+0.1 mg/L NAA,MS+3.0 mg/L 6-BA+0.2 mg/L NAA上再生不定芽,效果最佳,不定芽再生频率分别为72.7%,63.6%,69.2%。将不定芽切下转移至1/2 MS+0.5 mg/L IBA培养基上诱导生根,生根率达81%以上。The paper used the hypocotyls, cotyledon and cotyledon petiole as explants to study the effects of different hormone concentrations and combinations on induction callus and plant regeneration. The results indicated that the combinations of MS + 0.8 mg/L 2,4-D, MS + 0. 8 mg/L 2,4-D + 0. 1 mg/L 6-BA,MS +0. 8 mg/L 2,4-D +0.1 mg/L 6-BA as the induction callus medium was the best for hypocotyls, cotyledon, cotyledon petiole with 86.3% , 78.2% , 97.6% respectively of induction callus frequency, and then transferred callus of hypocotyls, cotyledon petiole, cotyledon to the optimal MS + 3.0 mg/L 6-BA + 0. 1 mg/L NAA, MS + 3.0 mg/L 6-BA + 0. 2 mg/L NAA medium for regenerating buds, in which the regenerating buds frequency was 72. 7% ,63.6% ,69. 2% respectively. The shoots were induced to form the 1/2 MS culture medium with IBA 0.5 mg/L at a frequency more than 81%.
