人IL－6在大肠杆菌中表达条件的研究

摘　　要：在完成了用基因重组技术把人白细胞介素６（ＩＬ－６）基因表达在大肠杆菌中的基础上，我们除了用不同的宿主菌，又用了不同的诱导时间及不同的培养基等条件对人ＩＬ－６表达的影响进行了进一步的比较研究。结果表明，人ＩＬ－６基因在Ｅ　ｃｏｌｌ　ＤＨ菌密度达ＯＤ６００　０．６５左右时，４２℃最佳诱导表达时间为５ｈ，其表达最高，ＳＤＳ－ＰＡＧＥ和凝胶密度扫描分析证明，表达的人重组ＩＬ－６（分子量２０．８ｋｕ）占菌体总蛋白的４１．０％，而在Ｅ－ｃｏｌｉ　ＲＲＩ及ＪＭ１０３中表达率最高值各为２４．７％（诱导４ｈ）和２２．４％（诱导５ｈ），用ＭｓＣＡ替代ＬＢ后，人ＩＬ－６在Ｅ　ｃｏｌｉ　ＤＨ５ｍ中的表达量降低很少。. coli DH5a/pH5a/pBVIL-6. a strain with the highest-level expression of human mature IL-6, wasobtained after a high effective human recombinant IL-6 (hrIL-6) expressing vector was transductedinto three strains of E. coli DH5a, RRI, JM103, and induced at 42℃ for 3～6 with molecu- lar weight 20. 8ku accounted for 41.0% of the E. coli proteins in DH5a (iduced for 5h). 24.7% inRRI (4h) and 22.4% in JM103 (5h). When the E. coli DH5a/pBVIL-6 was cultured and inducedwith the medium M9CA instead of LB, the amount of the hrIL-6 expression was almost not changed.which will be possibly very significant to the industrial preparation in the future.
