摘  要:目的:观察藻酸双酯钠(Polysccharide Su lfate,PSS)对培养神经细胞谷氨酸兴奋毒性的保护作用。方法:体外培养大鼠皮层神经细胞,加入谷氨酸观察谷氨酸对神经细胞的兴奋毒性及PSS的保护作用,以碘化丙啶染色,流式细胞仪定量分析细胞凋亡百分率;以荧光分光光度法测定细胞内游离钙及caspase 3活性的变化。结果:Pss50,100,150ug/m l能显著减少细胞死亡,降低乳酸脱氢酶(LDH)漏出量及丙二醛(MDA)生成,提高超氧歧化酶(SOD)活性,抑制细胞内[Ca2+]i累积以及caspase 3活性,抑制神经细胞凋亡。结论::PSS对培养神经细胞谷氨酸兴奋毒性具有显著的保护作用,其机制可能与PSS抑制细胞内[Ca2+]i累积以及抗脂质过氧化作用有关。Objective: To investigate protective effects of polysccharide on glutamate - induced neurotoxicity in cerebral cortical neuronal cultures. Methods: The Cortical neurons of fetal of rat were cultured in vitro. The protective effects of PSS on glutamate - induced neurotoxicity were observed. Apoptosis was measured via flow cytometry, the intracellular calcium and caspase 3 activity were determined by fluorescent assay. Results: PSS50, 100, 150μg ml^-1 could reduce glutamate - induced cell death, efflux of LDH , elevation of MDA , intracellular [ Ca^2+ ] accumulation and cell apoptosis as well as caspase 3 activity, in contrast, it increased the SOD activity. Conclusion: The protective effects on cerebral cortical neurons from ischemia injury have something to do with PSS, which can inhibit intracellular [ Ca^2+ ] accumulation and suppress generation of lipid peroxide.
