摘 要:考察了研磨法、有机溶剂法和超声波破碎法对根瘤土壤杆菌细胞破碎的影响。结果表明,在有机溶剂破碎条件下,最佳搅拌时间为3.5 h,此时CoQ10的含量为5.703 mg/L;由正交实验分析得出,菌体浓度为40 mg/mL,作用时间为12 min,破碎功率为250 W时,超声波破碎的效果最佳;与上述两种常规破碎方法比较,在研磨的条件下,加入抗氧化剂BHT(2,6-二叔丁基对甲酚)的细胞破碎效果最好,CoQ10的含量为8.012 mg/L,比不加抗氧化剂BHT研磨时提高了约38%,抗氧化剂BHT的最佳质量分数为20%。In this paper, the effects of milling , disruption with organic solvent and ultrasonic on disruption of Agrobacterium tumefaciens KY-3085(ATCC 4452) Strain were compared. The results showed that the yield of CoQ10 was 5. 703 mg/L when mixing time was 3.5 h, on the condition of disruption with organic solvent. The orthogonal experiment by ultrasonic cell disruption indicated the optimum conditions were 40 mg/mL concentration, 12 min, 250 W. Compared with above, milling with quartz and acetone was the best when the antioxidant BHT(2, 6-Di-tert-butyl-4-methylphenol) was added. In this method, the yield of CoQI0 was 8.012 mg/L, increased of 38 percent. The optimizated addition of antioxidant BHT was 20 %.
