加味瓜蒌薤白汤对实验性心肌缺血大鼠心脏保护作用的研究

摘　　要：目的观察加味瓜蒌薤白汤对实验性心肌缺血大鼠心电图、心脏病理显微结构、bax、bcl-2、Casep-ase-3、p53、VEGF含量的影响,探讨该方对实验性心肌缺血大鼠心脏的保护作用及机理。方法将Wistar大鼠80只随机分为空白对照组、模型组及全方高、中、低剂量组与全方去黄芪组、全方去黄芪加黄芪甲苷组、心得安组,各组分别灌胃2周造模,记录各组动物心电图;继续灌胃2周,取心脏行常规HE染色和SABC免疫组化染色,取血测VEGF。结果全方中剂量组、全方去黄芪加黄芪甲苷组与心得安组均可有效阻止异丙基肾上腺素引起的心肌断裂和心电图损害,与空白对照组相似。上述两组与心得安组均可通过抑制p53表达以调节bcl-2/bax的表达,并抑制Casepase-3的表达,抑制心肌细胞凋亡,与模型组相比差异有显著性。上述三组VEGF含量降低,与模型组比较差异有显著性;全方中剂量组及全方去黄芪加黄芪甲苷组与心得安组比较,差异亦有显著性。结论加味瓜蒌薤白汤治疗心肌缺血作用肯定,其作用机理与抑制p53表达,调节bcl-2/bax表达,抑制Casepase-3表达,抑制心肌细胞凋亡,降低VEGF含量相关。Objective:To observe the possible protective mechanism of Modified Gualouxiebai Decoction(加味瓜蒌薤白汤) on rats of experimental ischemia by studying the influence to Electro- cardiogram,microstructure,bcl- 2/Bas,P53,Csaepase- 3 and VEGF. Methods:Selected 80 Wister rats and distributed them into 8 groups at random:placebo contrast, model,high dosage,medium dosage,low dosage,dicoction without Huangqi(黄 芪 ),decoction with and propranolol group.Rats in each group were fed dividedly by 2 weeks,then recorded the ECG.Following,fed by another two weeks,killed the rats and dyed the hearts tissue with HE and SABC,at meanwhile took the blood for checking VEGF.Results:The astragalus medium dosage,deciction with astragalus and the propranolol could prevent the impairment of myocardium and ECG effectively caused by Isoprenaline,as similar as the placebo contrast.Contrasted with the model group,the groups of the medium dosage,decoction with astragalus and the propranolol can arrest notably in expression of P53,Casepase- 3,and improve notably in the expression of Bcl- 1/Bax.Compared with the model group,the groups of the medium dosage,decoction with and the propranlolo can lower notably the VEGF.Compared with the propranolol group,the groups of the medium dosage decoction with astragalus also lower notably the VEGF. Conclusion:The groups of the medium dosage,decoction with astragalus and the propranolol can stop the apoptosis of myocardial cells by arresting the expression of P53,Casepase- 3 and regulating the expression of Bcl- 2/Bax.Even more,the groups of the medium dosage,decoction with astragalus are more effective than the group of propranolol.
