摘 要:目的研究黄芪多糖对狂犬病疫苗免疫应答的影响。方法将BALB/c小鼠和昆明小鼠各自随机分为2组,即试验组与对照组,分别于0、7d接种冻干人用狂犬病疫苗(Vero细胞)。对照组疫苗用灭菌注射用水稀释,试验组疫苗用10%黄芪多糖(APS)溶液稀释,每只腹腔免疫0·5ml。BALB/c小鼠于15d无菌取脾,进行淋巴细胞转化实验;昆明小鼠在免疫前和第1针免疫后4、7、14、30、45和60d眶静脉采血,分别进行外周血淋巴细胞CTL试验和小鼠中和抗体水平测定。结果试验组疫苗诱导小鼠产生的中和抗体和细胞免疫水平均高于对照组疫苗,二者差异有显著意义。结论黄芪多糖能提高狂犬病疫苗的免疫原性。Objective To study the influence of Astragalus polysaccharide(APS) on the immtmogenicity of rabies vaccine for human use. Methods Divide BALB/c and Kunming mice into test and control groups and inject i.p. with freeze-dried rabies vaccine for human use, 0. 5 ml per mouse,on days 0 and 7 separately. The vaccine for test group was diluted with 10%Astragalus polysaccha- ride solution, and those for control groups with sterilized water for injection use. Take the spleen of BALB/e mice on day 15 for lympho- cyte transformation test ,and collect the venous blood of Kunming mice on days 4,7,14,30,45 and 60 for lymphocyte CTL test and determination of neutralizing antibody. Results Both the neutralizing antibody and cellular immune response levels of mice in test groups were significantly higher than those in control groups. Conclusion APS improved the immunogenicity of rabies vaccine for human use.
