摘 要:rFPV-IFNγS1是一株共表达传染性支气管炎病毒S1基因和鸡干扰素-γ基因的重组鸡痘病毒,将其接种SPF鸡3周后,用传染性支气管管炎病毒异源毒株攻击,考察重组疫苗对异源毒株攻击的保护作用。结果显示,疫苗接种后,免疫鸡血清中抗体很快产生,外周血中CD 4+和CD 8+T淋巴细胞与非免疫对照组相比都有显著差异。用LHB株IB病毒攻击后,免疫鸡的病理损伤明显轻于非免疫对照组,并且其排毒时间明显缩短,排毒量明显减少,攻毒后免疫组的发病率为30%(3/10),死亡率为10%(1/10),非免疫对照组的发病率为100%(10/10),死亡率为40%(4/10)。体重分析结果表明,攻毒前后免疫组与对照组相比,体重均没有显著变化,但攻毒后免疫组鸡体重增长的幅度要高于非免疫对照组。从以上结果可以说明,重组疫苗能在动物体内很好表达,对试验动物产生较好的保护作用,同时也减轻了重组疫苗对体重增长的抑制作用。rFPV-IFNγS1 is expressing ChIFNγ gene and IBV S1 gene recombinant fowlpox virus. In order to detect its protection to different strains, it was challenged with IBV LHB strain after immunized 3 weeks. The results showed that chickens in group immunized with rFPV-IFNγS1 produced ELISA antibodies soon. The ratio of CD8^+ and CD4^+ T lymphocytes in SPF chickens peripheral blood got notable changes compared to those in controls. Pathological slices of group immunized with rFPV-IFNγS1 didn't notable chage compared to those in controls and shorten the time of release virus and reduced the quantity of release virus. After challenging with the IBV LHB strain of group immunized with rFPV-IFNγS1, incidence was 30% (3/10), mortality was 10% (1/10), but incidence and mortality of control were 100% (10/10) and 40% (4/10). Weight analyzing results showed that group immunized with rFPV-IFNγS1 got no notable changes compared with control, but increasing extent of group immunized was higher than that of control. Taking all results together, rFPV-IFNγS1 recombinant vaccine was expressed very well in body, and it can protect SPF chickens against challenging with different IBV strain, and it can suppress the side effects by inoculating with recombinant live vaccine.
