摘 要:本研究探讨了乙二胺四乙酸钠(EDTA—Na)对猪卵母细胞的体外成熟和孤雌激活后早期发育的影响。利用屠宰场猪卵巢卵母细胞,在不添加或添加不同浓度EDTA—Na的成熟培养液中体外成熟44~48h,挑选核成熟卵母细胞进行孤雌激活,然后移到不添加或添加不同浓度EDTA—Na的胚胎培养液中进行体外培养168h。结果说明:(1)成熟液中添加适当浓度(25~100μmol/L)EDTA—Na对体外成熟猪卵母细胞孤雌激活后的早期发育具有促进作用,在EDTA—Na浓度为100μmol/L时,效果最佳;(2)胚胎培养液中添加适当浓度(25~100μmol/L)EDTA—Na,对体外成熟猪卵母细胞孤雌激活后的早期发育具有促进作用,在EDTA—Na浓度为100μmol/L时,效果最佳;(3)成熟液和胚胎培养液中同时添加100μmol/LEDTA—Na对体外成熟猪卵母细胞孤雌激活后的早期发育具有促进作用;(4)成熟液或/和胚胎培养液中添加不同浓度EDTA—Na,对体外成熟培养后的猪核成熟卵母细胞及孤雌激活后的4-细胞孤雌胚SDS—PAGE电泳的蛋白质表达图谱都无显著影响。This study was to investigate the effect of EDTA - Na on in vitro maturation of porcine oocytes and early development after parthenogenetically activation . Porcine oocytes from abattoir ovaries were matured in maturation medium whether or not containing different concentrations of EDTA-Na for 44~48 hours. Oocytes with the first polar body were parthenogenetically activated and then cultured in NCSU-23 medium whether or not containing different concentrations of EDTA-Na. The results showed: (1) Supplementation with 25~100μmol/L EDTA-Na in maturation medium was beneficial to early development of porcine parthenogenetic embryos derived from oocytes matured and artificially activated in vitro, and when EDTA-Na concentration was 100 μmol/L its effect was the best; (2) Supplementation with 25~100μmol/L EDTA-Na in embryo culture medium was beneficial to early development of porcine parthenogenetic embryos derived from oocytes matured and artificially activated in vitro, and when EDTA-Na concentration was 100 μmol/L its effect was the best; (3) Supplementation with 100μmol/L EDTA-Na in maturation and embryo culture media together was benificial to early development of porcine parthenogenetic embryos derived from oocytes matured and artificially activated in vitro; (4) Supplementation with different concentrations of EDTA-Na in maturation medium or/and embryo culture medium had no significant effect on protein expression profiles of SDS-PAGE electrophoresis of porcine oocytes matured in vitro and 4-cell parthenogenetic embryos after artificial activated.
