摘  要:目的:探讨肺癌、乳腺癌患者外周血中CD4+CD25+调节性T细胞水平的变化及临床意义。方法:应用流式细胞术检测9例肺癌、9例乳腺癌患者及10例非肿瘤患者外周血单个核细胞(PBMC)中CD4+CD25+T细胞占CD4+T细胞的比例,应用RT-PCR技术检测人外周血PBMCfoxp3基因的表达。结果:流式细胞术检测显示,肺癌及乳腺癌患者外周血PBMC中CD4+CD25+T细胞占CD4+T细胞的比例[(21·30±11·26)%]高于非肿瘤患者[(14·19±8·56)%](P<0·05),肺癌及乳腺癌患者外周血PBMC中foxp3mRNA的表达水平高于非肿瘤患者表达水平(P<0·05),肺癌和乳腺癌患者外周血CD4+CD25+调节性T细胞比例增加。结论:CD4+CD25+调节性T细胞在维持自身免疫稳定的同时对肿瘤免疫具有抑制作用,该细胞群比例的增加可能参与肿瘤的发生。Objective To investigate the changes of CD4^+CD25^+ regulator T cells in peripheral blood from the paitents with lung cancer and breast cancer. Methods Flow cytometry was used to analyze the proportion of CD4^+CD25^+ T cells/CD4^+ T cells in peripheral blood mononuclear cells (PBMC) from 9 patients with lung cancer, 9 patients with breast cancer and 10 patients without cancer as controls. The expression of foxp3 mRNA in PBMCwas detected by RT-PCR. Results The value of CD4^+CD25^+ T cells/ CD4^+ T cells in PBMC from patients with lung cancer and breast cancer (21.30± 11.26)% was significantly higher compared with controls (14. 19± 8.56)%. The expression of foxp3 in PBMC enhanced in patients with cancer compared with controls, there was significant difference between two groups (P〈0.05). It revealed that the population of CD4^+CD25^+ regulator T cells in peripheral blood of patients with cancer was increased. Conclusion CD4^+CD25^+ regulator T cells may be responsible for immune suppression in the tumor immunity while maintenance of immune tolerance and the increase of CD4^+CD25^+ regulator T cells may be involved in tumor generation.
