摘 要:目的研究头皮药物位点注射对缺氧缺血性脑损伤(HIBD)后脑细胞的修复作用。方法选40日龄Wistar大鼠32只,其中5只为正常对照,27只制成HIBD动物模型,20d后,随机分为A、B、C 3组,各9只。①A组:用VitB1及VitB12额顶叶处注射,1次/d,共25d。②B组:功能训练(Bobath法)。③C组:不处理。各组进行临床观察干预结束58d后处死动物,取脑组织进行光镜和电镜观察。结果①临床观察:B组与C组均有明显运动、记忆、行为异常和毛发改变;A组则无明显变化(见表1);②光镜观察:B组与C组均较A组神经细胞嗜酸性变严重,慢性变性神经细胞数量明显增多(P<0.05);B组又较A组严重,有显著性差异(P<0.05);B组与C组比较无明显差异(P>0.05)。③电镜观察:小血管的内皮细胞及星状细胞的损伤,A组均比B组和C组损伤轻(见图1-3)。结论HIBD模型可产生类似脑瘫的病理过程,药物注射疗法可明显改善神经细胞的损伤,产生修复作用。本试验为头皮药物注射治疗脑瘫提供一定的理论依据。Objective To study the repairing effects of epicranium medication injection treatment on brain cells of hypoxic-ischemic brain damage (HIBD) and its mechanism. Methods 32 Wistar rats of 40 day-old were selected, 5 of which were used as normal control group,27 rats were used as test animal models of HIBD. Twenty days later, they were divided into 3 groups randomly (n = 9). ①Group A was treatment group; the animals were treated with VitB1 and VitB12 by injecting over frontal lobe and apical lobe once a day for 25 days; ② Group B1 the animals were treated with Functional Training( Bobath methods) ;③Group C: no treatment. On the 7 days after treatment finished, all the animals in the three groups were killed and their brains were taken. The pathological changes of nerve cells, vascular endothelial cells, colloid cells and auxiliary fibers were observed under the light microscope and electronic microscope. Results ① Clinical observation: Animals in both Group B and C all had obvious dyskinesia, hypomnesia, behavior disorder and pelage changed, and those in Group A had no obvious change; ②Observed by light microscope: Group B and C all had more serious nerve cell eosinophil denaturation, and the amount of chronic degenerating nerve-cell increased obviously than those in group A. Group B have much more degenerating cells than those in Group A (P〈0.05). There is no significant difference between Group B and Group C(P〈0.05) ; ③ Observed by electronic microscope: The impairment on vascular endothelial cells and steller cells in thin blood vessels of albino rats in group A was much less than those in group B and C as was denoted. Conclusion HIBD model can form patho-process similar to Cerebral Palsy. Medication Injection Treatment relieve the injury of nerve cells and have better repairing effects. The experiment offered some theoretical evidence for Epicranium Medication Injection Treatment on Cerebral Palsy.
