摘 要:目的:探讨3种中草药滴眼液对体外培养的人癌细胞和正常细胞的细胞毒作用。方法:采用噻唑蓝(MTT法)测定鱼腥草滴眼液、葛根素滴眼液和双黄连滴眼液对6种细胞株(CNE2、G lc-82、HRPE、F ibro、3T3和ECV-304)的细胞毒作用并以半数抑制浓度(IC50)进行评价。结果:鱼腥草滴眼液对人癌细胞CNE2和G lc-82的IC50分别为11.5±0.25μl/m l和24.0±0.8μl/m l;对正常细胞HRPE、F ibro、3T3和ECV-304细胞的IC50分别为18.0±3.5μl/m l、52.0±14.0μl/m l、17.5±3.5μl/m l和17.5±1.3μl/m l。葛根素滴眼液对CNE2和C lc-82细胞的IC50为9.8±2.3μl/m l和17.0±5.0μl/m l,对ECV-304细胞的IC50为10.5±0.95μl/m l,而对HRPE、F ibro和3T3的IC50均>110μl/m l。双黄连滴眼液对CNE2、G lc-82细胞的IC50为18.9±5.0μl/m l和23.9±0.6μl/m l,对HRPE细胞的IC50为113.9±25.6μl/m l,对F ibro、3T3和ECV-304的IC50均>100μl/m l。结论:鱼腥草滴眼液对人癌细胞和正常细胞均有显著的细胞毒作用。葛根素滴眼液和双黄连滴眼液对正常细胞无明显的细胞毒作用。Objective : To study the cytotoxic effects of three kinds of eye drops on human cancer cells and normal cells. Methods : Cytotoxic effects of Yuxingcao Eye Drop ( YXC ) , Gegensu Eye Drop (GGS) and Shuanghuanglian Eye Drop (SHL) on 6 cell lines (CNE2, Glc-82, HRPE, Fibro, 3T3 and ECV-304) were assayed using MTT method. And cytotoxic efficacy was evaluated using fifty percent of inhibitory concentration ( IC50 ). Results : IC50 of YXC to human cancer cells CNE2 and Glc-82 were ( 11.5 ± 0.25 ) μg/ml and ( 24. 0 ± 0. 8 ) μt/ml ; to normal cells HRPE, Fibro, 3T3 and ECV-304 were ( 18.0 ± 3.5 ) μt/ml, (52. 0 ± 14. 0) μt/ml, ( 17.5 ± 3.5 ) μt/ml and ( 17.5 ± 1.3 ) μt/ml, respectively. IC50 of GGS to CNE2, Glc-82, ECV-304 cells were (9.8 ± 2. 3 ) μt/ml, ( 17.0 ± 5.0) μt/ml and (10.5 ±0. 95)μt/ml,respectively. Under the concentration of 100 μt/ml, the average survival rates of HRPE, Fibro and 3T3 cells were 60. 0% ,87.8% and 58. 2% , respectively. IC50 of SHL to CNE2, Glc-82 and HRPE cells were ( 18.9 ±5.0) μl/ ml, ( 23.9 ± 0.6 ) μl/ml and ( 113.9 ± 25.6) μl/ml. At the concentration of 100 μl/ml, the average survival rates of Fibro, 3T3 and ECV-304 Cells were 89.6% , 77.2% and 74.7%. Conclusion : YXC has evident cytotoxicities to human cancer cells and normal cells. While, GGS and SHL has not obvious cytotoxic effects to normal cells.
