百萨偃麦草高分子量麦谷蛋白亚基的鉴定及其染色体定位

摘  要:经SDS-PAGE电泳和W estern杂交,在百萨偃麦草(Thinopyrum bessarabicum)中鉴定出1个高分子量麦谷蛋白亚基,其电泳迁移率大于“中国春”小麦的1Dy12亚基,本研究命名为1Ebx亚基,其编码基因命名为Glu-1Ebx。对由百萨偃麦草衍生的双二倍体及其1套二体附加系进行SDS-PAGE分析,只有百萨偃麦草、双二倍体和1Eb附加系表达1Ebx亚基,小麦亲本及其它附加系均不表达此亚基。用1对HMW-GS通用引物对上述材料进行扩增时,只有在百萨偃麦草、双二倍体和1Eb染色体附加系的基因组DNA中扩增出分子量大小一致的DNA片段,与SDS-PAGE的定位结果一致。表明1Ebx亚基是由百萨偃麦草1Eb染色体上的HMW-GS基因所编码。A high molecular weight glutenin subunit (HMW-GS) which has a faster electrophoretic mobility than that of 1Dy12 subunit from the bread wheat Chinese Spring, was identified from Thinopyrum bessarabicum by SDS-PAGE and Western Blotting, designated as 1E^bx in this study. In order to determine the precise location of 1E^bx subunit, SDS-PAGE analysis was carried out in amphidiploid and additional lines deduced from Thinopyrum bessarabicum. Results of SDS-PAGE revealed that 1E^bX subunit was specially expressed in Thinopyrum bessarabicum, amphidiploid and 1E^b additional lines, other than wheat parent lines and other additional lines. Consistence with the location identified by SDS-PAGE, a band with the similar molecular weight was also amplified only in Thinopyrum bessarabicum, amphidiploids and 1E^b additional lines using a pair of degenerate primers of HMW-GS. These results showed that the 1E^bx subunit was encoded by a HMW-GS gene, Glu-1E^bx, on chromosome 1E^b of Thinopyrum bessarabicum.
