摘  要:探讨了一种新的筛选血细胞作用试剂的方法。通过对比表面活性剂十六烷基季胺盐(CTAB)、十二烷基聚氧乙烯硫酸钠(AES)和脂肪醇聚氧乙烯醚(AEO-9)对中性粒细胞的作用效果,为血细胞作用试剂的筛选提供理论基础。结果表明中性粒细胞经分离纯化后,表面活性剂对其作用可避免受到其他血细胞的影响,CTAB对红细胞的作用强于中性粒细胞,在较低浓度就能完全溶解红细胞,而对中性粒细胞的数量及大小改变不大;AES和AEO-9对红细胞的破坏能力比CTAB弱,不能将红细胞膜完全破碎,细胞膜碎片被计数为白细胞,干扰正常粒细胞计数分类;且低浓度AES作用粒细胞能减少其细胞数量,而AEO-9与CTAB随作用浓度增加,只是使粒细胞大小减少,不改变其数量。A new method is discussed to select a reagent reacting to blood cell. The experiments were made that cetyltrimethyl ammonium chloride (CTAB), sodium C12 - C14 fatty alcohol polyoxyethylene ether carboxylate ( AES ) and polyoxyethylene ( 9 ) fatty alcohol ether ( AEO-9 ) act on neutrophils. Comparing the results of the experiments an ideal method of selecting a reagent reacting to blood corpuscle were found. The results indicate that after the neutrophils are separated and purified, the effect of the surfactant on other blood corpuscle is avoided, so it reflects the exact effect caused by surfactant. And it was found that CTAB could completely lyse the erythrocytes with low concentration, and the quantity and volume of neutrophils have not be altered ; AES and AEO-9 couldn't completely lyse the erythrocytes, the erythrocytes ghost is counted as leukocytes, which consequently would disturb the counting of neutrophilics in whole blood, then the quantity and the percentage of neutrophils would be changed; AES with low concentration can decrease the quantity of granulocytes, with increasing concentration of AEO-9 and CTAB, the volume of neutrophils would be decreased, but the quantity of granulocytes wouldn' t be altered.
