摘  要:目的评价新型支架材料磷酸钙骨水泥-聚乳酸聚羟基乙酸二聚体(CPC-PLGA)复合物的生物相容性。方法噻唑蓝(MTT)比色法分析3种浓度材料浸提液(100%、50%、10%)对L929细胞生长的影响;分光光度法分析3种浓度材料浸提液的溶血性;复合物植入SD大鼠臀肌2周和4周后取出评价组织相容性;种植BMSC1周扫描电镜观察生长状况。结果24 h 3种浓度材料浸提液RGR分别为76%、76%、85%,48 h分别为75%、84%、96%,72 h分别为76%、77%、84%,细胞毒性评级均为1级;3种浓度浸提液对健康人血的溶血率分别为3.45%、2.59%、0.86%;肌肉内植入2周复合物组炎性反应为Ⅰ级4例、Ⅱ级5例、Ⅲ级1例,纤维囊腔形成10例均为Ⅳ级;4周复合物组炎性反应为Ⅰ级5例、Ⅱ级5例,纤维囊腔形成Ⅲ级3例、Ⅳ级7例;种植1周BMSC形态正常、生长状态良好。结论CPC-PLGA复合物无细胞毒性,无溶血作用,植入后无炎症反应,种植细胞生长良好,具有良好的生物相容性。Objective To evaluate the bioeompatibility of the new-type scaffold: CPC-PLGA polymer. Methods The effects of polymer extract with 3 concentrations (100%, 50% and 10% ) on L929 growth was detected by MTT colorimetry. The hemolysis of them was tested by speetrophotometer. Two and four weeks after the polymer was implanted into gluteus of SD rats, its tissue biocompatibility was analyzed. One week after BMSC were planted onto CPC-PLGA polymer, there interaction was observed under scanning electron microscopy. Results The RGR of polymer extract with 3 concentrations was 76% ,76% and 85% for 24 h,75% ,84% and 96% for 48 h,and 76% ,77% and 84% for 72 h,respectively. All of the grades of cytotoxicity were I. The hemolysis of polymer extract with 3 concentrations was 3.45 %, 2.59 % and 0.86 % respectively. The local inflammation of muscle was evaluated Ⅰ for 4 eases, Ⅱ for 5 cases, Ⅱ for Ⅰ ease,and the local fibre vesiea was evaluated IV for all 10 eases after 2 weeks;The inflammation of muscle was evaluated Ⅰ for 5 cases, Ⅱ for 5 cases,and the fibre vesica Ⅱ 3 cases, Ⅳ 7 cases after 4 weeks. BMSCs could grew on the material surface well for one week. Conclusion CPC-PLGA polymer had no cytotoxicity or hemolysis. It had no inflammation after transplantion. BMSCs could grew well on the surface of the material. CPC-PLGA polymer has excellent biocompatibility.
