摘 要:目的:探讨青藤碱在小鼠心脏移植中的抗排斥作用及其可能机制。方法:在小鼠心脏移植模型中,观察异系移植组、CsA治疗组、青藤碱治疗组移植心脏存活时间、术后7 d移植物排斥反应病理分级以及IL-2mRNA表达水平。结果:青藤碱治疗组移植心脏存活时间为(17.67±0.82)d,与异系对照组(11.00±2.10)d相比明显延长(P<0.01);病理结果显示,排斥反应减轻,呈轻-中度变化;移植心脏中IL-2mRNA表达水平明显减低(P<0.01)。结论:青藤碱在小鼠心脏移植中具有抗排斥作用,作用机制可能为抑制IL-2等促排斥细胞因子表达。Objective: To investigate the anti-rejection effect of sinomenine in mice heart transplantation and elucidate the anti-rejection mechanism of sinomenine. Methods: The mice with heart transplantation were divided into 3 groups: allograft transplantation group, CsA treatment group and sinomenine treatment group. The graft survival, pathological grading of graft rejection and IL-2mRNA expression levels on 7th day post-operatively were investigated in three groups. Results: The grafts survival was prolonged in sinomenine treatment group comparing with that in subgroup allograft transplantation (17.67 ±0.82 vs 11.00±2.10, P〈0.01).The rejection pathologically was relieved and mild to moderate changes appeared The IL-2 mRNA level in grafts significantly decreased in contrast (P〈0.01). Conclusion: Sinomenine possess anti-rejection effect in mice heart transplantation. The mechanism might be that sinomenine inhibits cytokine IL-2 which may promote the rejection.
