槲皮素对胃癌MGC-803细胞生长及凋亡作用的研究

摘　　要：目的：研究槲皮素对人胃癌MGC803细胞增殖的影响及诱导凋亡作用。方法：用四甲基偶氮唑盐（MTT）比色法检测不同浓度槲皮素对MGC-803细胞的增殖活性的效应；原位末端核苷标记（TUNEL）法检测槲皮素诱导细胞凋亡的凋亡指数（AI）。结果：槲皮素浓度为40～100μmol／L时对MGC803细胞增殖有显著的抑制效应，且呈剂量依赖性；细胞加入槲皮素诱导48h后AI明显高于未加入组（P〈0．05）。结论：槲皮素在一定浓度范围内能显著抑制MGC-803细胞的增殖并能诱导其凋亡．其作用呈浓度依赖性。OBJECTIVE： To study the effect of quercetin on the proliferation and apoptosis of human gastric carcinoma cell line MGC 803.METHODS： MTT assay was adopted to measure the effect of quercetin at different concentrations on the proliferation of MGC- 803 cells.Cell apoptosis index（AI） induced by quercetin was measured by TUNEL assay.RESULTS： Quercetin at a certain concentration from 40μmol/L to 100μmol/L could successfully inhibit the proliferation of MGC - 803 cells in a dose dependent manner.TUNEL assay indicated that apoptosis cells induced by quercetin in treatment group were much more than that in control group after 48 hours（P〈0.05） .CONCLUSIONS： Quercetin of certain concentration can evidently inhibit the proliferation of MGC 803 cells and induce their apoptosis with concentration dependent relation.
