摘 要:目的探讨小檗胺联合供体脾细胞输注诱导的微嵌合状态对皮肤移植物生存的影响。方法选择BALB/c(H-2d)、C57BL/6(H-2b)和615(H-2k)小鼠分别作为受体、供体和第三供体,采用不同的方式处理受体动物(小檗胺、供体脾细胞悬液输注、小檗胺联合供体脾细胞输注)诱导微嵌合状态,同时移植皮肤。观察皮肤移植物存活时间,逆转录-聚合酶链反应(RT-PCR)检测受体脾脏细胞因子(IL-2、IL-4)。流式细胞仪检测受体脾脏的微嵌合状态。结果小檗胺联合供体脾细胞输注可诱导微嵌合状态而形成免疫耐受,明显延长同种皮肤移植物的平均生存时间[MST=(26.5±7.0)d],排斥第三供体。实验组IL-2和IL-4均低于对照组与第三供体组。单纯小檗胺或供体脾细胞输注均可诱导微嵌合状态,也可延长皮肤移植物的生存时间[MST分别为(16.0±3.5)d和(12.7±2.1)d],但微嵌合状态的程度和延长皮肤移植物生存的作用不及小檗胺联合供体脾细胞输注(P<0.01)。微嵌合状态的程度与延长移植物生存时间无相关性。结论小檗胺联合供体脾细胞输注可诱导微嵌合状态产生免疫耐受。Objective To explore the effect of chimerism induced by donor splenocytes and berbamine on survival of mouse, skin allografts. Methods BALB/c ( H - 2d ) , C57BL/6 ( H - 2 b ) and 615 ( H - 2k ) mice were adopted as recipients, donors and the third part donors of skin allografts respectively. Treatment of the recipients with a course of berbamine, transfusion of a single dose of donor splenoeytes, a course of herbamine combined with infusion of viable donor splenocytes induced a ehimerism;meanwhile skin was transplanted onto recipients. Median survival time(MST) of transplanted skin allografts was used to evaluate the degree of transplantation tolerance,the semi - quantitative analysis of eytokiue gene expression( IL - 2 and IL -4 ) of spleens were determined by revere transcription polymerase chain reaction(RT-PCR) ,l:he level of chimerism in the recipient spleens was surveyed, Results MST of transplanted skin allogrdts was prolonged to ( 26. 5 ± 7. 0) days after berbamine administration and transfusion of donor splenoeytes while normally rejected the third part 615 skin grafts ( MST days) , ( 16. 0 ± 3. 5 ) days following berbamine injections, ( 12.7 ±2, 1 ) days behind infusion of donor spleen cells. In the control group,skin graftsMST was( 11, 3 ± 1.5 ) days, There was significant down - regulation of cytokine gene expression( IL - 2 and IL - 4) in the spleen in the berbamine combined with infusion of donor splenoeyte group, while the spleens in the control group and the third part donor skin grafts group had high levels of IL - 2 and IL - 4 gene expression. The chimeric degrees were different in variant groups, and were not correlated with the survival of skin grafts, Conclusion The treatment of berbamine combined with infusion of viable donor spleen cells may lead to a donor - specific tolerance,
