高效液相色谱法测定乳膏剂中环孢素的含量

摘　　要：目的：用高效液相色谱（HPLC）法测定环孢素乳膏剂中环孢素的含量。方法：色谱柱为VP-ODS C18柱（4．6mm×150mm，5μm）；流动相为乙腈-水-磷酸（760：240：0．5，v／v）；流速为1mL·min^-1；检测波长为210nm；柱温为60℃。结果：环孢素在20-400mg·L^-1范围内线性关系良好（r=0．9996），高、中、低浓度回收率（n=3）分别为98．6％（RSD为0．95％）、99．0％（RSD为0．82％）和99．2％（RSD为0．57％）。结论：该方法灵敏度高．线性范围广，具有较高的专属性，快速简便，重现性好，适合于该制剂的含量测定和质量控制。OBJECTIVE To establish an HPLC method for determination of the content of cyclosporin in its cream. METHODS The HPLC system consisted of C18 column（4. 6 mm× 150 mm, 5μm） with acetonitrile-water-phosphate （760：240：0. 5, v/v） as mobile phase, detection at 210nm, the flow rate was 1.0 mL,min-1 and column temperature at 60 ℃. RESULTS The linear range was 20 - 400 mg. L^- 1 （r = 0. 999 6）. The recovery （71 = 3） was 98. 6% at a higher concentration （with RSD of （1. 95%）, 99. （1%/40 at middle concentration （with RSD of 0. 82%） and 99. 2% at a lower concentration （with RSD of 0. 57%）. CONCLUSION The method is found to be simple, quick, accurate and suitable for determination of content of cyclosporin cream.
