佛波酯对大鼠移植肺的保护

摘　　要：目的应用两套管一支架管大鼠肺移植模型,观察佛波酯对大鼠移植肺的保护作用。方法分别应用缺血预适应以及外源性佛波酯同对照组相比,测定大鼠移植肺的左肺静脉氧分压,HSP70mRNA,SOD以及凋亡指数等指标,了解大鼠移植肺功能。结果缺血预适应组及佛波酯组的左肺静脉氧分压分别为(83.06±11.6)、(87.10±13.5)高于对照组(63.84±14.2);HSP70mRNA佛波酯组(1.504±0.43)和缺血预适应组(1.233±0.13)高于对照组(0.565±0.16),SOD佛波酯组(63.66±1.55)均高于预适应阻断组(51.85±15.2)和对照组(39.16±9.4)(P<0.05),而凋亡指数佛波酯组(16.50±4.88)、缺血预适应组(18.80±4.34)均低于对照组(25.00±6.99)(P<0.05)。结论大鼠供肺在移植前,经外源性佛波酯灌注后,能够增强HSP70mRNA的表达,进而提高SOD的活力,具有肺保护作用。Objective Using two - cuff - one - stent lung transplantation model, to investigate the effects of phorbol myristate acetate （PMA） on transplanted lung in rats. Methods 30 SD rats were randomly divided into three groups： control group （the donor lung was preserved normally）, preconditioning group （the donor lung underwent ischemic preconditioning before preservation） and PMA group （phorbol myristate acetate was infused via pulmonary artery to take place of ischemic preconditioning）, respectively. PO2 of left pulmonary vein, HSP70mRNA , SOD activity and apoptotic index were detected when the donor lung were anastomosed. Results All the parameters of PO2 [ （ 83.06 ± 11.6）,（87.10±13.5）vs（63.84 ±14.2）], HSP70mRNA[（1.504±0.43）,（1.233±0.13）vs （0. 565±0. 16） ] ,SOD（63.66±1.55） and apoptotic index in preconditioning group （51.85 ± 15.2） and PMA group were significantly higher than those in control group（39. 16±9.4） （ P 〈0. 05）while apoptotic index [ （ 16. 50 ± 4. 88 ）, （ 18. 80 ± 4.34 ） ] are all lower than that of control group （ 25.00 ± 6. 99）, （ P 〈 0.05）. Conclusion Phorbol, which was infused before the lung preserved, can strengthen the expression of HSP70mRNA , and improve the activity of SOD. Phorbol can mimic the protective effects of ischemic preconditioning.
