摘 要:目的观察加味四逆散对大鼠恶劣心境障碍模型纹状体单胺类神经递质的影响。方法选择Open-Field法评分相近的50只成年SD雄性大鼠,随机分为对照组、模型组、氟西汀组和加味四逆散大、小剂量组,每组10只。应用分养和长期不可预见性中等强度应激造成大鼠恶劣心境障碍模型。以HPLC-EC法对照研究恶劣心境障碍大鼠纹状体单胺类神经递质的改变。结果与对照组比较,模型组大鼠纹状体3-甲氧基-4-羟基苯乙二醇(MHPG)、去甲肾上腺素(NE)、多巴胺(DA)、3,4-羟基苯乙酸(DOPAC)、5-羟吲哚乙酸(5-HIAA)和5-羟色胺(5-HT)的含量下降。与模型组比较,氟西汀组MHPG、NE、5-HIAA和5-HT含量升高;加味四逆散大、小剂量组MHPG、NE、DA、DOPAC、5-HIAA和5-HT含量升高。结论恶劣心境障碍模型大鼠纹状体单胺类神经递质含量降低,加味四逆散对恶劣心境障碍的治疗作用可能与改善纹状体单胺能神经的功能活动有关。Objective To investigate the effect of Jiawei Sini powder on monoamine neuro transmitters in striatum of rat dysthymic disorder Model. Methods Male Sprague-Dauley rats were randomly allocated to five groups as follows: control, model, Fluoxetine, Jiawei Sini powder Ⅰ (general dose) and Jiawei Sini powder Ⅱ (overdose). The rat dysthyrnic disorder model was produced by separation and chronic unpredictable mild stress. The behavior of rat was detected by open-field test. HPLC-EC was used to study the changes of monoamine transmitters in striatum of rat dysthyrnic disorder model. Results MHPG, NE, DA, DOPAC, 5-HT and 5-HIAA in the model rats were lower than control. Compared with the model, MHPG, NE, HVA, 5-HT and 5-HIAA in the Fluoxetine were higher, MHPG, NE, HVA, DA, DOPAC, 5-HT and 5-HIAA in the Jiawei Sini powder Ⅰ and Jiaweisini powder Ⅱ were higher. Conciusion The monoamine transmitters in striaturn of rat dysthymic disorder model are decrease and Jiawei Sini powder is effective in the treatment of dysthymic disorder by regulating the function of monoamine transmitters in sriarum.
