摘  要:目的检测谷氨酰胺转运体ASCT2、LAT1及LAT2在豚鼠耳蜗中的表达,初步探讨谷氨酸-谷氨酰胺循环中,谷氨酰胺从支持细胞进入内毛细胞的机制。方法选取健康白色红目豚鼠6只,Trizol法提取耳蜗中的总RNA,逆转录聚合酶链反应检测ASCT2、LAT1及LAT2在耳蜗中的表达。结果正常豚鼠耳蜗中有ASCT2、LAT1的表达,且ASCT2表达较强,LAT2在耳蜗中无表达。结论在正常豚鼠耳蜗中含有谷氨酰胺转运体,支持细胞中的谷氨酰胺可能是通过谷氨酰胺转运体进入内毛细胞中。OBJECTIVE To detect the expressions of ASCT2,LAT1 and LAT2 in guinea pig cochlea and explore their effects for the mechanism of glutamine was transported from supporting cells into inner hair cells during glutamate-glutamine cycle. METHODS The total RNA of guinea pig cochlear was extracted by Trizol one step method and the expressions of ASCT2,LAT1 and LAT2 in cochlear were detected with reverse transcription polymerase chain reaction. RESULTS The expressions of ASCT2,LAT1 were detected in guinea pig cochlear, and the expression of ASCT2 was stronger than LAT1, while there was no expression of LAT2 . CONCLUSION The exsistence of glutamine transporters in guinea pig cochlear suggested that glutamine in supporting cells could be transported into inner hair cells through glutamine transporters.
