熊胆贝母止咳胶囊的体外抗病毒作用的实验观察

摘  要:目的:观察熊贝止咳胶囊是否具有体外抗病毒的作用。方法:按常规病毒体外试验,首先将熊贝止咳胶囊粉溶于Hank'S液,以二倍稀释法配制成1:20~1:2560浓度,分别加入狗肾传代细胞(MDCK)和Hela细胞培养板中,100μl·孔^-1,连续7日于3TC5%CO2孵箱中培养,逐日观察,测其对细胞无毒性作用的最大药物浓度。然后以100TC赋的流感病毒川属99和柯萨奇病毒(CVB3)分别加入MDCK和Hela细胞培养板中,分别吸附2.Oh和1.5h,去病毒液后,于MDCK细胞培养板孔中分别加入熊贝止咳胶囊最大无细胞毒性药物浓度稀释液,并以病毒唑1:20~1:20480作平行阳性对照。而在Hela细胞培养板中,分别加入无细胞毒性熊贝止咳胶囊1:80~1:640的药液,亦以病毒唑1:80~1:640作平行阳性对照。100μl·孔^-1,每浓度3孔,并设空白细胞和病毒对照,37℃5%CO2孵箱中培养,逐日观察细胞病变,当病毒对照组细胞出现约有75%以上细胞病变时终止试验,当细胞发生约50%细胞病变以上时,则判受试药物该浓度对该病毒无抑制作用。结果:测得熊贝止咳胶囊对MDCK和Hela细胞的无毒性浓度分别为1:2560和1:80;而病毒唑对两株细胞的无毒性浓度均为1:20。实验显示熊贝止咳胶囊1:2560-1:20480的各种浓度对流感病毒川属99无抑制作用;1:80~1:640的浓度对CVB3无抑制作用。结论:熊贝止咳胶囊对MDCK和Hela细胞的无毒性浓度以下各浓度,对流感病毒川属99和CVB3在体外无明显的抑制作用。Objective:To investigate the antivirus activity of Xiongbei antitussive(XBA) capsules. Methods: 1, The powder of XBA capsules were double diluted continually to 1: 20- 1 : 2560 concentration solutions with Hank's balanced salt solution. And then adding 100(1 of the solution to each microtiter plate which was planted with Madin-Darby Canine Kidney( MDCK) cell or Hela cell. Culturing the microtiter plates in 5% CO2 incubator at 37℃ for 7 days. Observing and recording the cell toxic reaction caused by drug daily for determining the drug's maximal non-toxic concentration to cells. 2, Adding 100 TCID50 of the inflnenza virus (Sichuan genus 99) and Coxsackie virus (CVB3) into MDCK and Hela cell microtiter plates. After adsorbed in the plates 2. 0 hours for the inflnenza virus and 1.5 hours for CVB3, getting rid of all the virus solutions. Adding the maximal non-toxic concentration of XBA capsule solution to MDCK cell into MDCK cell microtiter plates and using the ribavirin 1 : 20-1 : 2480 as the parallel positive control group. In hela cell plates, adding the 1 : 80-1 : 640 XBA capsules" maximal non-toxic concentration and using the ribavirin 1 : 80-1 : 640 as the parallel positive control group as well. All the holes of the plates were added 100(1 of the drug solution and every different concentration of the solution had 3 holes repeated including control and positive control groups. 4, Culturing the microtiter plates in 5% CO2 incubator at 37℃. Observing and recording the cell toxic reaction caused by drug every day. Then ending the experiment when virus positive control group cell appeared 34+ about 75% cells had pathological changes). Considering the concentration has no inhibit effect on the virus when cell appeared 2+( about 50 % cells had pathological changes). Results: The XBA capsules ; maximal non-toxic concentration to MDCK cell and Hela cells were 1 : 2560 and 1 : 80 respectively. However the ribavirin's maximal non-toxic concentration to M
