CⅡ变构肽对胶原性关节炎T细胞免疫的影响

摘  要:目的探索连续替换CⅡ多肽中268-270位氨基酸的变构肽对胶原性关节炎(CIA)抑制作用的可能机制。方法CⅡ268-270变构肽90μg静脉注射,每周1次治疗CIA大鼠;设置无关肽对照组和空白对照组。ELISA方法检测大鼠血清TH1/TH2细胞因子水平及抗CⅡ抗体及其亚型。荧光实时定量RT-PCR检测调节性T细胞标记Foxp3 mRNA表达。结果CⅡ268-270变构肽、无关肽及空白对照组血清IFN-γ水平分别为41.42±7.32、54.63±11.15和57.86±5.64 pg/ml,变构肽可降低大鼠血清IFN-γ水平(P<0.01)。变构肽、无关肽及空白对照组血清IL-10水平分别为40.18±5.13、31.09±5.04和30.19±5.02 pg/ml,变构肽可以提高大鼠血清IL-10水平(P<0.01)。变构肽、无关肽及空白对照组抗CⅡIgG2a抗体滴度分别为0.5±0.21、1.1±0.27和1.3±0.29,变构肽可以显著降低抗CⅡIgG2a抗体滴度(P<0.01)。变构肽、无关肽及空白对照组抗CⅡIgG1抗体滴度分别为1.2±0.24、0.58±0.11和0.6±0.12,变构肽可以显著上调抗CⅡIgG1抗体滴度(P<0.01)。变构肽、无关肽及空白对照组Foxp3 mBNA表达丰度分别为33.62±12.12、10.05±6.08和9.73±6.03,变构肽可以显著上调大鼠Foxp3 mRNA表达丰度(P<0.01)。结论CⅡ268-270变构肽可以调节CIA中异常的TH1/TH2细胞因子平衡,可以抑制IgG2a型抗CⅡ抗体产生,诱导Foxp3阳性T细胞产生。Objective To explore the mechanism of the inhibitory effect of CⅡ268-270 altered peptide on collagen-induced arthritis. To study its effect on TH1/TH2 balance, anti-CⅡ IgG antibody isotypes and regulatory T celia. Methods The altered peptide was given intravenously with a dose of 90μg once a week. Peptide control and blank control were treated with similar approach. The serum TH1/TH2 cytokines were measured by ELISA. The anti-CⅡ antibody and its isotypes were measured with ELISA. The induction of regulatory T cells were assayed by real-time RT-PCR analysis of its marker Foxp3. Results The serum IFN-γ in rats treated with altered peptide, control peptide and blank control were 41.42±7.32, 54.63±11.15 and 57.86±5.64 pg/ml respectively (P 〈 0.01). The serum IL-10 in rats treated with altered peptide, control peptide and blank control were 40.18±5.13, 31.09±5.04 and 30.19±5.02 pg/ml respectively (P 〈 0.01). The Foxp3 mRNA abundance in spleen mononuclear cells of rats treated with altered peptide, control peptide and blank control were 33.62±12.12, 10.05±6.08 and 9.73±6.03 respectively (P 〈 0.01). Conclusion CⅡ268-270 altered peptide could regulate the balance of TH1/TH2 cytokines. It could decrease anti-CⅡ IgG2a antibody. It could also up-regulate Foxp3^+ T cells in vivo.
