摘 要:采用3因素3水平L9(34)的正交设计,以组织块增殖百分率作为评价指标,研究了在M199和RPMI-1640培养基中分别添加Ca2+,Mg2+,Zn2+金属离子对鲍的外套膜和上足触手组织培养的影响.结果显示,Ca2+和Zn2+对细胞增殖具有明显的促进作用,Mg2+的作用不明显;对上足触手细胞生长最好的组合为5 g/L的Ca2+,1 g/L的Mg2+,20μg/L的Zn2+;对外套膜细胞生长最好的组合为1 g/L的Ca2+,1 g/L的Mg2+,60μg/L的Zn2+.The effects of three metal ions on the cultured tissue from mantle and epipodial tentacle of Haliotis discus hannai with Ca^2+ , Mg^2+ and Zn^2+ ions which are added into the M199 and RPMI- 1640 medium are studied respectively. The experiment is projected according to orthogonal design L9 (3^4), the proliferation percent of tissue blocks is estimated as the indicator of growth.The results show that the Ca^2+ and Zn^2+ have an obvious effect on accelerating the proliferation of cultured cells, but the effect of the Mg^2+ is not evident. When Ca^2+ is 5g/L, Mg^2+ is 1g/L and Zn^2+ is 20μg/L, the growth of cells of epipodial tentacle is growth conditions of the mantle cells are that the concentration of Ca^2+ is 1g/L, Mg^2+ 1g/L the concentration of the best. But the best and Zn^2+ 60μg/L.
