摘  要:为了提高昆明白小鼠MⅡ期卵母细胞的去核效率及质量,该研究在不同条件下,比较了小鼠卵母细胞的去核效果:将小鼠MⅡ期卵母细胞放入含蔗糖浓度分别为0.01、0.02、0.03 g/mL的M2-液微滴中,在体式显微镜下观察显核效果以确定最佳浓度;将有清晰可见细胞核的卵母细胞放入含细胞松弛素B(Cytocha-lasin-B,CB)浓度分别为5、7.5、10μg/mL操作液中去核,显微镜下确定去核率。结果表明:操作液中加入蔗糖浓度为0.02g/mL时显核效果最好;去核时操作液中细胞松弛素B浓度为10μg/mL时去核死亡率最低。To increase the effect of mice oocytes enucleation, in the study, the influence of different conditions on the effect of enucleation was compared: MⅡ oocytes were put in M2- culture medium respectively containing 0.01,0.02,0.03 g/mL sucrose, then karyon was observed by solid microscope. The optimal concentration would be ascertained. The oocytes that their karyon can be observed were put in operation medium respectively containing 5,7.5,10 μg/mL CB and enucleated. The rate of enucleation can be noted. The results show that : The operation medium containing 0.02 g/mL sucrose make for observing karyon; When the operation medium containing 10 μg/mL cb, oocytes mortality is minimum.
