摘  要:目的探讨P 2Y 12基因多态性与冠心病(CHD)的相关性。方法177例CHD患者为观察组,61例健康成年人为对照组。应用PCR-RFLP技术检测两组P 2Y 12基因C 34T、G 52T突变基因型。结果观察组C 34T多态位点T突变等位基因频率显著高于对照组,ACS与SAP间比较无统计学意义。T等位基因与CHD正关联。G 52T多态等位基因频率和基因型频率在两组间分布差异均无统计学意义。结论CHD患者存在P 2Y 12基因多态性;T突变型等位基因可能具有提高个体对CHD的易感性及个体凝血系统活化水平的作用。[Objective] To examine the distribution of P2Y12 gene polymorphisms in Chinese population and the correlatio of these polymorphisms with atherosclerotic heart disease (CHD). [Methods] Genotypes and frequencies of two sites were examined by polymerase chain reaction -restriction fragment length polymorphism technique in 61 unrelated normal control individuals and 177 CHD patients. [Results] The frequency of T allele in CHD patients was significantly higher than that in controls. When the patients were divided into acute coronary syndrome (ACS)and stable angina pectoris(SAP), the frequency of T allele in ACS patients was higher than that in controls, there was no significant difference between ASC patients and SAP patients. While the frequency of T allele in SAP patients was higher than that in controls. There was a positive correlation between T allele and CHD. The distributions of G52T allelic and genotype frequencies did not significantly differ between CHD patients and controls. [Conclusion] T allele for site 34 of P2Y12 gene may has the positive effect on the susceptibility of CHD and high level of coagulation response.
