摘  要:LM-3是一株来自土壤的广谱拮抗细菌.经革兰氏染色、形态特征、培养特征和生理生化特征等分析,LM-3菌株被鉴定为多粘类芽孢杆菌.采用盐析法提取获得了该菌株的抗菌粗蛋白,抑菌活性测定表明抗菌蛋白对辣椒炭疽病菌等多种植物病原真菌有强烈的抑制作用.抗菌蛋白对链霉蛋白酶E和胰蛋白酶不敏感,对蛋白酶K部分敏感;高温(120℃)处理20min后抗菌蛋白仍保留84.71%的活性;酸碱处理发现抗菌蛋白在pH 8.5时最为稳定,而在pH 2.5时80℃处理1h完全失活;25W紫外灯(2400lx)照射8h对其活性没有影响.发酵上清液经硫酸铵盐析、高温(100℃)处理和SDS-PAGE分析,该抗菌蛋白的分子量介于6kDa和14.2kDa之间.The broad-spectrum antagonistic bacterium strain LM-3 was isolated from soil. This environmental isolate was determined to be PaenibaciUus polymyxa based on its Gram reaction, morphological characteristics, growth characteristics and biochemical characteristics. Antifungal protein produced by this strain was obtained by precipitation, It was shown on PDA plates that the development of many pathogens such as Colletotrichurn gloeosporioides was inhibited by the antifungal protein effectively. It was found to be tolerant to Pronase E and trypsin and partially sensitive to proteinase K; Aftertreated with high temperature (120℃) for 20 minutes, the antifungal protein still reserved 84.71% of its antagonistic activity; It was steady at pH 8.5 but lostall its activity at pH 2.5, 80℃ for an hour; It was tolerant under the treatment of UV(24001x, 8h). The antifungal protein was precipitated with (NH4)2SO4 from cell-free filtrates derived from cultures of strain LM-3, then treated with high temperature ( 100℃ ) and analyzed by SDS-PAGE electrophoresis; the results showed that its molecular weight was between 6kDa and 14.2kDa.
