摘 要:目的:观察针刺对N-甲基-N-亚硝脲诱导的大鼠视网膜变性视网膜的保护作用。方法:N-甲基-N-亚硝脲以50 mg/kg剂量腹腔注射于7周龄的SD大鼠诱导视网膜变性,然后随机分为针刺治疗组、模型对照组,观察针刺治疗7 d后两组大鼠视网膜的形态学变化,并与同一时间点的正常组进行比较。结果:N-甲基-N-亚硝脲引起的大鼠视网膜变性明显,针刺组大鼠全视网膜外核层的损伤程度轻于模型组,外核层光感受器细胞的核固缩、深染以及光感受器的内、外节的损伤均轻于模型组。结论:针刺能够抑制N-甲基-N-亚硝脲诱导的大鼠视网膜光感受器细胞的病理损伤。Objective: To observe the impact of acupuncture on morphological changes of retinal cells after intraperitorsi injection of N-methyl-N-nitrosourea (IVlNU) in the rat. Methods: Twenty 50-day-old female SD rats were randomized into control ( n = 4), model ( n = 8), and acupuncture ( n = 8) groups. Retinal degeneration model was established by intraperitoneal injection of MNU (50 mg/kg) in the rats of the later two groups. Bilateral "Xirrning" (behind the ear, the mid-point of the folding region inferior to the ear's lower angle), and "Jingning" (BL 1 ) were punctured once daily, continuously for 7 days. Following finishing the experiments, the rat's eyeballs were taken out to be immersed in 4% paraformaldahyda, embedded in paraffin, sectioned, stained with hematoxylin and eosin stain, and observed by using light microscope and transmission electron microscope (TEM). Results: Compared with normal control group, the outer nuclear layer (ONL) thickness and the total thickness of the retina in model group showed thinning significantly (P〈 0.01 ), and the cells' arrangement was in disorder; when compared with model group, the ONL thickness and the total thickness in acupuncture group were markedly bigger (P〈 0.01 ) and the cellular arrangement was relatively well-arranged. Results of TEM showed smaller (withered) cellular body, condensed nuclear chromatin, swelling of the mitochondria with severer vacuole, etc in model group, while in acupuncture group, the apoptosis of the photoreceptor cells was apparent but the injured degree was much lighter than that of model group. Conclusion: Acupuncture can protect retinal photoreceptor cells from MNU-induced injury in rats.
