摘 要:目的探讨甘草总黄酮体内抗肿瘤作用并阐明其可能的作用机制。方法建立S180小鼠肉瘤,观察甘草总黄酮对S180小鼠肉瘤抑瘤率,肿瘤组织切片HE染色观察其病理改变,从肿瘤组织提取的DNA并做琼脂糖凝胶电泳,用免疫组化法检测瘤组织内Bcl-2和Bax蛋白的表达水平。结果甘草总黄酮各组能显著抑制S180小鼠肉瘤生长,肿瘤组织HE染色观察,部分肿瘤细胞体积较小,细胞核浓染。肿瘤组织的DNA电泳带呈明显的梯状条带;免疫组化显示小鼠肿瘤组织内的Bcl-2蛋白的表达下调,而Bax蛋白的表达上调。结论甘草总黄酮体内有明显的抑瘤作用,抑瘤机制可能与其诱导肿瘤细胞凋亡密切相关,影响肿瘤细胞内相关凋亡蛋白Bcl-2和Bax蛋白的表达实现的。Objective To study the anti-tumor effects of the total flavonoids from radix glycyrrhizae in vivo, and to explore its possible mechanisms of the anti-tumor effect of the total flavonoids from radix glycyrrhizae. Methods The mouse tumor model S180 was used to investigate the effects of the total flavonoids from radix glycyrrhizae on tumor growth, sections from transplanted tumors in mice was subjected to HE staining to reveal the changes of histomorphologism; and the DNA of tumor tissue was extracted for agarose gel electrophoresis , the expression of Bcl-2 and Bax protein in tumor tissue were detected using inmmnohistochemistry assay. Results The total flavonoids from radix glycyrrhizae had obvious effects on tumor inhibition, Hematoxylin-eosin (HE) stain showed that cell volume was reduced , nuclear chromatin was condensed and cytoplasm was stained deeply with red under light microscope, the agarose gel electrophoresis of the DNA showed ladder figure. The expression of Bcl-2 protein was down-regulated, while expression of Bax protein was up-regulated in tumor tissue treated with the total flavonoids from radix glycyrrhizae compared with the control groups. Conclusion The results suggest that the total fiavonoids from radix glycyrrhizae has obvious anti-tumor effect in vivo. It is likely to induce the tumor cells apoptosis.
