中国人庚型肝炎病毒部分基因的克隆及序列分析

摘　　要：目的：分析中国人血清中庚型肝炎病毒（ＨＧＶ或ＧＢＶ－Ｃ）ＮＳ３区部分基因序列。方法：从个体供血者及肝硬化患者血清中，通过逆转录合成ｃＤＮＡ，设计特异性引物进行多聚酶链式聚合反应，将产物克隆于载体上，采用双脱氧核苷酸链终止法进行双向测序。结果：分离出的５株ＮＧＶ毒株的ＮＳ３区部分核苷酸序列之间同源性从８５％到９８％，与国外已发表的ＨＧＶ序列比较，同源性在７９％～９１％。推测的其编码氨基酸序列，５株之间１００％一致，与国外已报告的ＨＧＶ比较，同源性为９４．９％～１００％。结论：从中国人血清中分离出５株ＨＧＶ序列，其ＮＳ３区部分核苷酸存在着一定的变异性，但其编码的氨基酸非常保守。Objective:To clone and sequence the partial genome of hepatitis G virus(HGV or GBV-C)in the serums from the Chinese,Methods:Total nucleic acids were extracted from the serums of paid donors and liver cirrhosis patients and subjected to reverse transcriptase PCR using the primers derived from the putative NS3/ helicase region.The PCR products were cloned into the vectors and were se-quenced by double-strand DNA sequence analysis kit.Results:Among the five isolates (HGV-C1～C5),the nucleotides identity is in the region from 85% to 98% and the deduced amino acids are all the same.Compared with the published HGV isolates,the nucleic acid sequences of HGV-C1～C5 are 79%to 91% identical and the deduced amino acid sequences exhibit 94.9%～100% homology.Conclusion:Five Chinese isolates of HGV were obtained.The local nucleotide sequences of the putative NS3 region of HGV have a visible variability,but the amino acid sequences encoded hy these nucleic acids are highlyconserved.
