摘 要:为评价中药“病毒克”颗粒剂体外抗新城疫病毒(NDV)的活性,采用细胞培养技术,通过细胞病变抑制法和MTT比色法检测“病毒克”颗粒剂在鸡胚戍纤维细胞(CEF)中对新城疫病毒增殖的影响。结果表明“病毒克”颗粒剂对CEF的半数中毒质量浓度(TC50)为61.35mg/mL,抗NDV的半数有效质量浓度(EC50)为3.2mg/mL,治疗指数(TI)为19.2,“病毒克”颗粒剂对新城疫病毒具有直接灭活作用和阻断其感染CEF的作用,并存在明显的量效关系(P〈0.01);在感染后9h内以14mg/mL剂量给予“病毒克”颗粒剂,对新城疫病毒复制的抑制率为42.8%~76.596。说明“病毒克”颗粒剂对新城疫病毒在鸡胚成纤维细胞中的增殖具有明显的抑制作用。The objective of this study was to observe the effect of Bingduke granules (Chinese medicinal herb) on Newcastle disease virus (NDV) in vitro. Cytopathic effect inhibition and MTT colorimetric assays were used to detect the anti-viral activity of Bingduke granules on NDV in chick embryo fibroblast (CEF) cells, The median toxic concentration (TC50), median efficacious concentration (EC50) and treatment index (TI) were 61.35 mg/mL, 3.2 mg/mL and 19.2, respectively. The NDV was inactivated directly and the function of NDV infection in CEF was blocked by Bingduke granules in a concentration- dependent manner (P 〈 0.01). Bingduke granules could effectively inhibit the replication of NDV when added to CEF cell within 9 hours at the dosage of 14 mg/mL after infection. These results show that Bingduke granules has significant inhibitory effect on NDV replication in vitro.
