摘 要:目的了解天津地区夏季一起小学校流感样暴发疫情中分离出2株甲1亚型流感病毒株HA1基因变异情况。方法MDCK细胞和鸡胚双腔法分离流感病毒,收获病毒液提取病毒的RNA,进行RT-PCR,扩增产物纯化后测序,用DNASTAR软件进行序列分析。结果新分离株HA1基因长度为325个氨基酸,与国际疫苗株A/NewCaledonia/20/99相比氨基酸同源性高于98%,氨基酸替换位点有5个,其中165位位于抗原决定簇Ca1区。结论天津地区新分离甲1亚型流感病毒抗原性发生进一步漂移,但变异程度不大。Objective To understand the genetic characteristics of the newly isolated influenza A(H1N1) virus in a influenza outbreak in Tianjin. Methods The influenza virus was isolated by MDCK cells and embryonated eggs. Virion RNA extracted by Rneasy RNA extraction kit was used for RT-PCR amplification of the gene HA1. Therafter, the purified PCR products were sequenced. The data obtained were analysed with the software DNASTAR. Results The H1N1 viruses isolated were 325 amino acids, the homogeneity of amino acids, as compared with A/New Caledonia/20/99 strain, was 98% higher, and have 5 different amino acids on HA1 protein molecule ,165 position was in antigenic determinant Cal. Conclusion Little genetic changes occurred in the HA1 domain of the new isolated influenza A viruses(H1N1) in Tianjin.
