摘 要:目的探讨炒紫苏子醇提物对5-脂氧酶(5-LO)的抑制作用。方法大鼠胸腔内注入致炎物质(角叉菜胶),制备多形核粒细胞悬液,超声破碎,得到5-脂氧酶粗提物,设阴性对照组、紫苏子12.8,25.6和51.2 mg.L-1、木犀草素5组,ELISA法检测LTB4释放水平;体外观察炒紫苏子醇提取物对5-LO和15-LO酶活性的抑制作用。结果12.8,25.6和51.2 mg.L-1炒紫苏子醇提取物及木犀草素能够明显抑制胸腔多形核粒细胞悬液LTB4释放(P值均<0.01),抑制率分别达到75.06%,79.04%,81.26%,81.43%;炒紫苏子醇提取物对5-LO酶活性具有明显的抑制作用,其IC50为79.73 mg.L-1。结论紫苏子醇提取物能够抑制5-脂氧酶酶活性。OBJECTIVE To study the inhibitory action of EtOH extraction of masted pefilla seed(RPS) on 5-lipoxidase activity. METHODS Cawageenan was injected into the pleural space of rats, the rats were killed by decapitation at 5 h, PMNs were harvested from the pleural cavity and the activity of 5-lipoxidase was determined. RESULTS The production of the LTB4 from the PMNs was decreased significantly at the dose of 12.8, 25.6 and 51.2 mg·L^-1 EtOH extraction of RPS, and at the dose of 0.36 mg·L^-1 luteolin, compared with the negative control( P 〈 0.01 ). Their inhibition percentages were 75.06 %, 79.04 %, 81.26 % and 81.43 %, respectively. 5- Lipoxidase activity was inhibited by the EtOH extraction of RPS with the IC50 of 79.73 mg·L^-1. CONCLUSION The EtOH extraction of masted perilla seed can inhibit 5-lipoxidase activity.
