摘  要:目的探讨外用维甲酸脱色素作用的机制。方法利用体外培养的melan-a黑素细胞,观察全反式维甲酸和阿达帕林对中波紫外线(UVB)诱导的melan-a黑素细胞黑素合成的影响。结果二者可剂量依赖性抑制UVB诱导的细胞酪氨酸酶活性和黑素合成,使其回复到基础水平,但对基础黑素合成无明显影响,10-5M的全反式维甲酸可抑制黑素细胞增殖,阿达帕林对细胞增殖无明显影响。全反式维甲酸和阿达帕林对细胞黑素合成的影响无明显差异。结论提示维甲酸可直接作用于黑素细胞抑制其黑素合成,这可能是维甲酸脱色素作用的机制之一。Objective Retinoic acid(RA) is a hormone-like agent involved in the control of cell differentiation. Several clinical studies have demonstrated that topical tretinoin improves mottling and hyperpigmented lesions of photodamaged skin. The basic mechanisms underlying this effects of RA are not known. To determin whether RA could directly modulate excessive melanin synthesis, we investigated the in vitro effect of all-trans-retinoic acid and adaplen on UVB-induced melanogenesis in melan-a mouse melanocytes. Methods Cultured melan-a melanocytes were irradiated by UVB radiator, followed by treated with all-trans-RA and adaplen separately, and then melanin content and tyrosinase activity were measured. Results The two RAs in a dose-dependent manner significantly decreased the UVB-stimulated melanogenesis in terms of tyrosinase activity and melanin synthesis, which returned to basal levels. However, there was no significant modification in basal melanogenesis in melan-a mouse melanocytes in the presence of RA only. The two kinds of RAs elicited the same response concerning on their effects on melanogenesis. AU-trans-RA had moderately inhibitory effect on cellular proliferative poten- tial, the inhibitory rate was 45% at the concentration of 10-SM, while adaplen slightly but not significantly inhibited celluar proliferation. Conclusion Our results suggested RA inhibite melanogenesis directly in melan-a mouse melanocytes, which may explain partly the mechanisms of depigmentation by RA.
