摘 要:目的:观察南虎凝胶剂对实验性创伤性滑膜炎兔滑膜组织中NF-κBp65表达的影响,以探讨南虎凝胶剂治疗创伤性滑膜炎的作用机制。方法:采用电针刺激新西兰兔的股四头肌和腓肠肌,引起兔膝关节长时间被动屈伸而导致关节滑膜损伤来制作创伤性滑膜炎的动物模型。分5组给药治疗,5d后处死动物,对动物膝关节滑膜组织病理切片后进行免疫组化染色,观察NF-κBp65阳性细胞情况以了解各组动物关节滑膜组织NF-κBp65的表达水平。结果:模型组NF-κBp65的表达比正常组显著升高(P<0.01);而预防组、低剂量组和高剂量组的NF-κBp65表达水平明显降低,与模型组比较差异有显著性(P<0.05),其中高剂量组与模型组比较差异更有显著性意义(P<0.01)。高剂量组、预防组与正常组比较NF-κBp65的表达水平差异无显著意义(P>0.05),但低剂量组与正常组比较NF-κBp65表达水平差异有显著意义(P<0.05)。结论:南虎凝胶剂有抑制滑膜组织NF-κBp65表达的作用,减轻了兔关节创伤性滑膜炎炎症,并且剂量越大效应越显著,从而推断南虎凝胶剂对于创伤性滑膜炎可能有良好的预防和治疗效果。Objective To observe the effect of Nanhu gelatin on the expression of NF-kappa Bp65 in synovial membrane tissue of experimental synovitis induced by trauma and to explore its mechanism of action in the treatment of synovitis. Methods We established an animal model of traumatic synovitis through stimulating New Zealand rabbit's quadriceps femoris and gastrocnemius with an acusector to cause repeated passive movement of their knee joints leading to synovial membrane injury. The rabbits were divided into 5 groups based on the experimental design and were executed 5 days after Nanhu gelatin therapy. The levels of NF-kappa Bp65 expressions in the synovial membrane tissue of knees joints in all groups were detected after the synovial membrane tissues were stained by immunohistochemistry. Results NF-kappa Bp65 expression in the model group was significant higher than that in the normal group (P 〈 0.01) while NF-kappa Bp65 expressions in the prevention and low-dose groups were markedly decreased than that in the model group (P 〈 0.05) and NF-kappa Bp65 expression in the high-dose group was decreased more significantly (P 〈 0.01). NF-kappa Bp65 expressions in both high-dose and prevention groups were no more significant than that in the normal group (P 〉 0.05) whereas there was significant difference in NF-kappa theBp65 expression between the low-dose and the normal group (P 〈 0.05). Conclusion Nanhu gelatin can reduce inflammation of the rabbit's impaired synovial membrane in a dose-dependent manner by inhibiting the expression of NF-kappa Bp65, which indicates that it may be used to prevent and treat traumatic synovitis.
