摘 要:目的:研究胃缺血-再灌注损伤(gastric ischemia-reperfusion injury,GI-RI)对大鼠胃窦黏膜D细胞内生长抑素(somatostatin SS)含量的影响。方法:实验组大鼠分为单纯缺血组(ISO)、GI-R1 h、6 h、24 h和72 h组(I/R1-4),用免疫组织化学法及图像分析法对SS进行定量研究。结果:正常大鼠胃窦黏膜中D细胞主要分布于幽门腺的中、下1/3处,形态多样;而ISC组和I/R1组D细胞多为圆形,很少见有长的胞质突起;在I/R2-4组,则可见D细胞形态多样,并可见分布于幽门腺上1/3处。ISC组和I/R1组D细胞的数目显著少于对照组,细胞平均灰度值极显著大于对照组;而I/R2-4组D细胞的数目均显著多于对照组,仅I/R2组细胞平均灰度值极显著低于对照组。结论:胃窦黏膜中D细胞的形态数量及细胞内SS的含量在大鼠GI-RI过程中有变化,提示SS参与了GI-RI过程。Objective. To explore the effect of gastric ischemia-reperfusion injury on somatostatin content in the rat antral gastric mucosa D cells. Methods: Rats were divided into ischemia (ISC) and ischemia-reperfusion groups. The animals were killed at 1, 6, 24 and 72 hours after reperfusion separately. The somatostatin contents in the rat antral gastric mucosa D cells were observed by using the immunohistochemical method combined with image analysis technique. Results. D cells were predominantly present in middle lower zone of the pyloric glands in control rats with various shapes. In IS(2 and I/R1 groups, most D cells were round, with few protrusion. In I/ R2-4 groups, D cells were various in shape and were also observed in upper zone of the pyloric glands. The numbers of D cells in the IS(; and I/R1 groups were significantly less than those of control group, while the average grey density of D cells was significantly higher than that of control group. In I/R2-4 groups, the numbers of D cells were more than those of control group markedly; the average grey density of D cells in I/R2 group was significantly lower than that the control group. Conclusion: There are alterations of morphology and number of D cells and somatostatin contents in the rat antral gastric mucosa in response to ischemia-reperfusion, suggesting that somatostatin is involved in the process of gastric ischemia-reperfusion injury.
