摘 要:目的:观察苦参碱注射液对老年痴呆(Alzheimer’s disease,AD)模型大鼠脑组织中IL-1β含量及海马神经元超微结构的影响。方法:采用鹅膏蕈氨酸(ibotenic acid,IBO)双侧海马立体定位注射造模,用苦参碱注射液治疗30d后,放射免疫法测定IL-1β含量;电镜观察大鼠海马神经元超微结构的变化。结果:模型组与空白组比较,大鼠大脑皮质和海马内IL-1β的含量非常显著升高;高剂量治疗组与模型组比较,大鼠大脑皮质和海马内IL-1β的含量显著降低,且大鼠海马神经元线粒体超微结构的损伤有较大改善。结论:苦参碱注射液能降低大鼠脑内IL-1β的含量,从而保护线粒体结构,改善能量代谢,延缓神经元凋亡,进而控制AD的发展。Objective: To study the effect of Matrine injection on the level of IL-1β and ultrastructural changes in the central nervous system of the rats injected with ibotenic acid (IBO). Methods: IBO was directly injected into hippocampus of rats; Matrine injection was administered intraperitoneally for 30 days (treatment group). The level of IL-1β was assayed with radioimmunoassay and ultrastructural changes in hippocampus of the rats was observed with transmission electron microscope. Results: The levels of IL-1β in the frontal cortex and hippocampus were significantly higher and there were mitochondria swelling and cristae fragmentation. Synaptic corpuscle decreased in model group compared with those of the control group. After Matrine injection administration, the IL-1β contents in the frontal cortex and hippocampus were decreased and ultrastructural changes of neuron were improved. Mitochondria swelling and cristae fragmentation, synaptic corpuscle were decreased compared to the model group. Conclusion: Matrine injection can decrease the level of IL-1β in the AD rats caused by the IBO and improve the pathological ultrastructural changes of the central nervous system.
