MTX对大鼠胶原型关节炎免疫学机制的研究

摘　　要：目的探讨氨甲喋呤(MTX)治疗大鼠胶原型关节炎的免疫学机制。方法健康SD大鼠,用牛Ⅱ型胶原诱导产生胶原型关节炎(CIA),CIA大鼠经MTX肌注治疗4周后,观察各组大鼠关节炎指数,ELISA方法测定大鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)及抗牛Ⅱ型胶原抗体的含量。结果与对照组相比,肌注MTX能明显改善大鼠胶原型关节炎症状(P<0.05);CIA大鼠肌注MTX后,血清中TNF-α含量(60.6±21.3)pg/ml和抗牛Ⅱ型胶原抗体的OD值水平(0.68±0.07),分别与对照组(246.1±29.3pg/ml和0.81±0.04)相比明显降低(P<0.05);肌注MTX的CIA大鼠血清中IL-10含量(33.4±9.8)pg/ml与对照组相比(4.6±1.5)pg/ml明显升高(P<0.05)。结论MTX能有效地降低CIA大鼠血清中TNF-α和抗Ⅱ胶原抗体的水平,同时提高IL-10的含量。Objective To explore the immulogical mechisms of treating rat＇s collagen ＂arthritis（CIA） with methotrexate （MTX）. Methods Collagen arthritis was induce in healthy SD rats with cattle＇ s type 2 collagen . CIA rats were treated with MTX intramuscularly for 4 weeks before arthritic index were recorded , TNF- α, IL- 1, IL- 4, IL- 10 and type Ⅱ Anti - Bovine collagen antibody was accessed by ELISA. Results ①compared wi.h the control group , MTX administered intramuscularly could significantly release the symptoms of rat＇ s collagen arthritis（CIA） （ p 〈 0.05 ） ; ②the contents of TNF - α（60.6 ± 21.3 pg/ml ）and OD value of type Ⅱ Anti - Bovine collagen antibody （0.68 ± 0.07 ）in serum of CIA rats reduced significantly compared with those in control group （246.1 ± 29.3 pg/ml and 0.81 ± 0.04 ） （ p 〈 0.05 ） ; ② serum level of IL - 10 in rats of CIA administered with MTX intramuscularly （33.4 ± 9.8 pg/ml）increased significantly compared with those in control group （4.6 ± 1.5 pg/ml ）（ p 〈 0.05 ）. Conclusions MTX could effectively decrease serum TNF - α and type Ⅱ Anti - Bovine collagen antibody value in rats of CIA and increase serum level of IL- 10 at the same time .
