摘  要:目的:观察RAD16-Ⅱ的物理特性以及神经干细胞在其上黏附和增殖的情况,探讨其构建的细胞假体在脊髓损伤中可能的应用。方法:将RAD16-Ⅱ的饱和水溶液同与之等体积的1.8%的氯化钠溶液混合后静置24h,通过肉眼、光镜和扫描电镜观察其表面结构,并对其吸水性和溶液pH值的动态变化进行评价;将经鉴定的神经干细胞与其共同培养,扫描电镜下观察细胞在材料上的黏附情况,同时应用免疫组化方法评价神经干细胞在材料上的增殖和分化情况。结果:RAD16-Ⅱ在1.8%的氯化钠溶液中塑型后,肉眼和光镜下形态多变,其形状取决于塑型的容器形状;扫描电镜下其为规则的网状,网孔直径为54.00±3.24"m,纤维直径为9.00±1.57"m,网孔底部相互沟通,神经干细胞可在其上迁移和增殖。结论:RAD16-Ⅱ具有良好的物理特性,神经干细胞可在与其构建的假体中迁移和良好地增殖、分化,在脊髓损伤的修复中可能具有潜在的应用前景。Objective:To investigate the characteristics of RAD16-Ⅱ and its adherence and proliferation in heron stem cell,and its roles in spinal cord injury.Method:RAD16-Ⅱ were dissolved in the same volume of 1.8% NaCl solution.The face of RAD16-Ⅱ was observed by SEM and microscope and its hydrophilia and changes of value pH in its solution was analysed.Neural stem cells (NSCs) attaching to RAD16-Ⅱ to form cellular prostheses were identified.Then adhereance and migration of neural stem cells on the RAD16-Ⅱ were observed by SEM and cellular proliferation was evaluated by dyeing nucleus with DAP1 (4',6-diamidino-2- phenylindole).Result:After RAD16-Ⅱ were shaped in 1.8% NaCl,their faces were seemed as regular net under SEM with the diameter of mesh of 54.00±3.24μm and diameter of fibra of 9.00±1.57μm.Neural stem cells' migrate and proliferate on the RAD16-Ⅱ scaffold was documented.Conclusion:Neural stem cells attaching to RAD16-Ⅱ pertained with wonderful characteristic which make their proliferation,differentiation and migration as normal.The cellular complex will be widely used in SCI repair.
