肾安提取液对3T3细胞增殖及α-肌动蛋白表达影响的实验研究

摘　　要：目的:从细胞学方面探讨肾安提取液阻缓肾纤维化的作用及其机理。方法:采用3T3细胞系作为成纤维细胞模型,以碱性成纤维细胞因子(bFGF)为刺激因子,分别进行细胞增殖和免疫组化实验。结果:肾安提取液对3T3细胞的抑制作用呈显著的剂量依赖关系;48 h 6.25 mg/m l,12.5 mg/m l,25 mg/m l组、72 h 3.12 mg/m l,6.25 mg/m l,12.5 mg/m l,25 mg/m l组与对照组对细胞增殖的影响比较,差异有统计学意义(P<0.05,P<0.01);其中25 mg/m l组72 h与48 h比较,差异有统计学意义(P<0.05);但与尿毒清组比较差异无统计学意义(P>0.05)。肾安提取液对bFGF刺激的α-肌动蛋白(α-actin)表达的抑制作用随剂量增加而增强。结论:提示肾安提取液可能通过抑制bFGF刺激的成纤维细胞的增殖及α-actin表达,从而减缓肾纤维化。Objective： To study the effects and mechanisms in inhibiting renal fibrosis of Shen＇an extraction from cellular level. Methods： 333 cell was adopted as the model of fibroblasts with base fibroblast growth factor（bFGF） as the stimulated factor. On the cell multiplication and immunity tissue chemical experiments we are currently working on. Some results from our project include： The inhibitive effects of Shen＇an＇s extraction of the 333 cell took on a obvious dose - dependent relationship, In Shenhn extraction group with the concentrations of 6.25 mg/ml, 12.5 mg/ml, 25 mg/ml（ treated for 48 hours）, 3.12 mg/ml, 6.25 mg/ml, 12.5 mg/ml, and 25 mg/ml （treated for 72 hours）. The differences had a statistical significance in cell multiplication compared with the control group （P 〈 0.05, P 〈0.01 ） ; so was 25 mg/ml （treated for 72） hours compared with 48h. , but it＇s statistical significance were insignificant compared with Liaoduqing＇s group （P 〉 0.05 ）. It was enhancement with a dosage greater than the inhibitive effects of Shen＇an＇s extraction on the expression of α - actin stimulated by bFGF. Conclusion ： The result indicated that Shen＇an＇s extraction could slow down the renal fibrosis possibly through inhibiting the cell multiplication and the expression of α -actin stimulated by bFGF.
