摘  要:目的探讨葛根素对慢性脑供血不足(CCCI)病人的临床疗效及作用机制。方法选择CCCI病人96例,予葛根素400mg静脉输注,每日1次,连续14d,分别于治疗前后用TCD测量脑血流速度和阻力指数(RI),用比色法测定血浆丙二醛(MDA)、超氧化物歧化酶含量(SOD),用放免法测定血浆内皮素(ET)、降钙素基因相关肽(CGRP)水平。结果葛根素治疗2周后:头晕、头重、失眠总有效率分别为97.9%、96.0%、71.9%;大脑中动脉和椎动脉血流速度增快,血管RI下降;血浆MDA和ET含量较治疗前下降(P<0.01),而SOD和CGRP水平较治疗前上升(P<0.01)。结论葛根素治疗CCCI安全有效,能降低MDA、ET含量,提高SOD活力和CGRP的释放。Objective To study the effect and mechanism of Puerarin for chronic cerebral circulation insufficiency (CCCI). Methods Puerarin was injected with CCCI in a dose of 400 mg/day for 14 days in 96 patients, The peak of velocity (VP), resistance index (RI) of middle cerebral artery (MCA) and vertebral artery (VA) were studied with TCD. The levels of plasma malonic aldehyde and superoxide dismutase (SOD) were determined with chromatomety, the levels of endothelin (ET) and calcitonin gene- related peptide(CGRP) were determined with radioimmunological assay(RIA) pre- and post - therapy. Results Following Puerarin treatment for 14 days, the total effective rate for dizziness, top - heaviness and insomnia were 97.9%, 96.0% and 71,9% respectively. VPs of MCA and VA were faster post- therapy than that pre- therapy(P 〈 0.01 ). Plasma MDA, ET and Rls were lower post - therapy than that pre : therapy (P 〈 0.01 ). SOD and CGRP were higher pos - therapy than that pre - therapy ( P 〈 0.01 ). Conclusion Puerarin was very effective for CCCI. Puerarin could reduce the content of plasma MDA and ET, increase the activity of SOD as well as promote the release of CGRP.
