通心络胶囊对大鼠血管球囊损伤后胶原增生的影响

摘　　要：目的通过大鼠颈总动脉球囊损伤模型,观察通心络胶囊对大鼠血管球囊损伤后血管内膜胶原增生的影响。方法动物随机分为假手术组、手术组、通心络大剂量组及小剂量组,每组8只。假手术组、手术组均术后6周予蒸馏水3mL/(kg·d)灌胃,通心络大剂量组予通心络1.0g/(kg·d)灌胃,通心络小剂量组予通心络0.5g/(kg·d)灌胃。用ELISA法检测各组术后6周I型前胶原羧基肽(PIP)、氧化低密度脂蛋白(ox-LDL)的水平,病理形态学观察细胞增殖情况,计算机图像分析内膜、中膜厚度及其比例。结果动脉球囊损伤后6周,药物治疗组与手术组比较,可抑制内膜增生、降低内膜/中膜厚度、降低PIP、ox-LDL含量(P<0.05)。结论通心络胶囊对大鼠血管球囊损伤后血管内膜胶原增生有一定的抑制作用。Objective To observe the effects of Tongxinluo Capsule on collagen proliferation of artery after balloon injury in rats. Methods Thirtytwo healthy male SD rats were divided randomly into four groups： Sham operation group （ n = 8）, operation group （ n = 8）, low - dose group （ n = 8） [Tongxinluo Capsule 0. 5 g/（ kg · d） ] and high - dose group （ n = 8 ） [Tongxinluo Capsule 1.0 g/（kg· d）]. Following the management of animals, the pathomorphological examination was carried out and intimal thickness was analyzed by the computer imaging analysis system. The PIP and ox- LDL level were measured by ELISA. Results The intimal thickness, the PIP and ox-LDL levels in the operation group were lower than that in the medical treatment groups. Conclusion Tongxinluo Capsule had preventive effect on collagen proliferation of artery after balloon injury in rats. It might be effective for prevention and therapy of angioplasty- induced restenosis.
